The theory of stress has gradually become a kind of new theory of integral physiology and pathology, since 1936, Hans Selye defined it as a specific psychological and physiological change occurred in human body based on the term "stress" in the filed of physics. The research about stress plays an important role in the domain of health and diseases of human being. With the transformation of modern medicine mode, it shouldn't be ignored that many negative stimulants still do harm to human body. As the mechanism of stress is uncertain, no effectual drug can counteract its pathological damage in western medicine to date. The research about the essence of the Liver's function in Chinese medicine in recent years indicates that its physiological function and pathological change closely relate with the neuroendocrine disorders and immunity dysfunction etc induced by stress. Therefore, it is significant to explore the mechanism and treatment strategy of stress in light of Chinese medicine and pharmacology.Two parts including theoretical and experimental study were involved in this thesis which explored the mechanism of stress and action of Chinese herbs on it based on the theory of Chinese medicine.Theoretical study1. Stress Stress is a series of neuroendocrine reaction of human body and varieties of functional and metabolic change which ensues from that caused by varieties of severe stimulating factors (stressors). The activation of hypothalamic-pituitary-adrenocortical axis is important characteristics of stress reaction with the complicated mechanism. Hippocampus is involved in integrating the cognition information, expounding the meaning of environmental information, modulating behavior and neuroendocrine reaction. Many stressors can substantially increase the plasma β-endorphin. Endo-opium system and its receptors play an important role in modulating stress. Sympathetic-adrenal medulla system and hypothalamic-pituitary-adrenocortical hormone change and so do immunity function in the meantime under the circumstance of stressors. The neuro-endocrine-immunity network exerts on the regulation of stress reaction.2. Relation of stress with the Liver in Chinese medicine The Liver's function in Chinese medicine closely correlates with neuro-endocrine disorders and immunity dysfunction in the state of stress of body. The Function that Liver is in charge of soothing and regulating in Chinese medicine plays an important role in thestress of body. The Liver regulates the stress reaction through its impact on the motion of Qi, blood and emotions.3. Mechanism of Chinese medicine for stressKnowledge about stress and mechanism of stress reaction are attributed to change of the motion of Qi in the theory of Chinese medicine. It is thought that the disorder of Qi and blood and Zang-fu dysfunction induced by the disorder of the motion of Qi is one of main reasons that cause stress-related diseases. Anti-stress of Chinese herbs mainly takes effect through regulating Yin-yang, Qi, blood and Zang-fu.Experimental studyAny stress reaction is a process which is divided into three stages: vigilance, resistance and exhaustion in the angle of adaptation. Simple rat model induced by immobilization stress was applied in this experiment to observe multiple indices including animal behavior and neuro-endocrinology etc and explore the regulation effect of recipes of soothing-liver Chaihushugansan and Xiaoyaosan on stress. The results were as follows.1. General observation: Rats in model group were lassitude in appearance, gregarious and reluctant to move, no gloss in their hairs and less water and fodder in-take accompanied with loose stool. The eight-gaining in the normal group slowed down and the fodder intake was much less than that in the control group.2.
