Soothing The Liver And Invigorating The Spleen And Promoting Blood Circulation Recipe On Liver Tissue In Rats With Nonalcoholic Steatohepatitis FAT/CD36 Expression

Objective:to observe representative prescriptions of the therapies of soothing the liver,tonifying the spleen and invigorating the circulation of blood:CSGC(Chaihu shugan powder),Erchen decoction combined with Siling Powder,Gexiazhuyu decoction on hepatic FAT/CD36 expression in rats with nonalcoholic fatty liver,to discuss the possible mechanisms to prevent NASH with different.TCM therapies.Methods:NASH model in rats were induced with 12 weeks of high-fat diet,meanwhile all the model rats were treated with 12 weeks of 6g·Kg-l·d-1 of Chaihu shugan powder?14 g·Kg-1·d-1 of Erchen decoction combined with Siling Powder?14g·Kg-1·d-1of Gexiazhuyu decoction.the hepatic pathologic change in rats were observed with Oil red O and HE staining.The hepatic TG,CHOL and serum transaminase were detected with the biochemical method,the expression of hepatic FAT/CD36mRNA,TNF-?mRNA and IL-6 mRNA were detected with Realtime PCR method.The hepatic FAT/CD36 protein expression was detected with immunohistochemical method,the serum and hepatic TNF alpha and IL-6 were detected with ELISA.Results:(1)the hepatic fattv degeneration.inflammatory necrotic foci in hepatic lobule,hepatocyte ballooning and the sores of NAFLD activity in the model were obviously higher than in the normal group.the serum ALT and AST and hepatic TG,CHOL vwere significantly higher than in normal group(P<0.05.P<0.01);the inflammatory necrotic foci in hepatic lobule.NAS.serum ALT,AST and hepatic TG,CHOL in rats of the treatment groups were obviously,lower than in the model group.(P<0.05,P<0.01).(2)the expression of hepatic CD36,TNF-a and IL-6 gene mRNA and protein in model group were significantly enhanced than in the normal group(P<0.05.P<0.01),serun TNF-a and IL-6 were significantly higher than in the normal group(P<0.05.P<0.01):the CD36.TNF-a and IL-6 mRNA and protein expression in the treatment groups were significantly decreased than in the model group(P<0.05.P<0.01),while Serum TNF-a and IL-6 were significantl)decreased than in the model group(P<0.05.P<0.01)Conclusion:(1)CD36 takes part in the development of NASH induced by high-fat diet.(2)the therapies of soothing the liver,tonifying the spleen and invigorating the circulation can regulate the liver lipid metabolic disorder,reduce liver inflammation and prevent from the progress of NAS by means of inordinate of inhibition of CD36.
