Cerebrovascular disease is a kind of common disease in clinic. The incidence rate and fatality rate heads the list of successful candidates in domestic or foreign area. Among them, The incidence rate of ischemic cerebrovascular disease is far more than hemorrhagic cerebrovascular disease. With the development of modern medicine, the means and methods to cure ischemic cerebrovascular disease increased. The problem of ischemia-reperfusion is involved in the procedure of nature or therapy. So, it is very important to study ischemia-reperfusion in brain and to find operative means and methods of how to control it in mordern medicine. It is thousands of years passed of acupuncture to treat cerebrovascular disease. This is a good chance for us to solve the problem by acupuncture.Many pathologic mechanism is full of the course of brain ischemia-reperfusion injury. The cascade of damage is one course of brain ischemia-reperfusion injury. It includes four parts: excitotoxicity, periinfarct depolarization, inflammation, programmed cell death. TGFβcan increase the product of bcl-2 and decrease the concentration of Ca2+ in cells. So it can protect the nerve cells from toxic effect and oxidating response of glutamic cells. TGFβ can inhibite inflammation. TGFβcan increase the expression of transcription factor as c-jun, jun-B, c-fos and increase the plasticity of nerve cells. TGFβ can accelerate the regeneration of blood vessel and preclude the no-reperfusion in ischemia-reperfusion injury. It can recover the supply of blood in the ischemical area. TGFβcan prevent the damage of nerve cells in ischemical reperfusion injury. GAP-43 can make G0 and GTP-γ-s compound.This makes the passage of Ca2+ in cells closed and decrease the concentration of Ca2+ in cells and stop the damage of nerve cells. GAP-43 can influence the expression of nerve growth factor (NGF) and accelerate the regeneration and plerosis of nerve cells. GAP-43 is the sign of regeneration of mature nerve cells. We can infere the effect and degree of the means and methods to the rats of focal brain ischemia-reperfusion injury by observing the change of TGFβand GAP-43.In this experiment we selected wistar rats. We made model of focal brain ischemia-reperfusion injury by middle cerebral artery occlusion (MCAO). Ischemia is 2 hours and reperfusion is 72 hours. We observed the expression of GAP-43 in the cryosection of rats brain and TGFβcontent in serum of rats after 72 hours of reperfusion. We observed the influence to them of acu, medicine (naofukang) and compound of acu and medicine (naofukang). So we can discuss the nerve protection mechanism of acupuncture to focal brain ischemia-reperfusion injury and provide reliable evidence for acupuncture to control brain ischemia-reperfusion injury in clinic.Results:There is almost not positive cells of GAP-43 in the cryosection of normal controlling group. We can see a few positive cells of GAP-43 in the section of sham-operated group. That means weak expression in them. It has notable defference with the normal controlling group (P<0.01). The number of GAP-43 positive cellsin model group is far more than normal controlling group or sham-operated group (P<0.01, P<0.01). The number in medicine group is notable defferent with model group (P<0.05). This shows that medicine (naofukang) has some function to brain ischemic-reperfusion injury. The number in acu group is defferent with model group (P<0.05). The number in acu group is not notable defferent with medicine group (P>0.05). This shows that acu has some function to brain ischemia-reperfusion injury. The number in compound acu and medicine group is notable defferent with model group (P<0.01). The number in compound acu and medicine group is defferent with medicine group or acu group (P<0.01, P<0.01). This shows there is synergistic function to brain ischemia-reperfusion injury of acu and medicine.The level of TGFβis very low in rats serum of normal controlling group.
