The large vessels disease is the main reason to causing death and mutilation of the 2 type diabetes mellitus patients, a serious of investigations of pathology find that arteriosclerosis large vessels disease of 2 type diabetes mellitus can begin before blood sugar increase. In addition, the traditional arteriosclerosisthe risk factors, such as lipoi-demia, hypertension, cant explain risk of cerebrovascular accident in IRS patients, which hint there are other agents play roles, therefore, under the state of insulin resistance, the alteration of function of vessel endothelial cell attracts the researchers of domestic and abroad.NO and ET -1 are blood vessel active factor which are produced by vessel endothelial cell, both of them regulate the vasodilation and vasoconstriction, the proliferation of smooth muscle cells maintains the normal function of endothelial cells.This experiment set up a model of insulin resistance through high - fat and high - fructose diet feeding, comprehend the alteration of vessel endothelial cells indirectly and explore the abnormal relationship between it and lipo - metabolism by the estimating the marker of NO and ET -1. We expect that it can offer a new way to prevent the large vessels disease and better the related diseased prognosis.Materials and Methods60 male adult healthy Wistar rats, 200 - 250grams, were dividedinto A, B, C groups random, which stand for control group, high - fat diet group and high - fructose diet respectively. The A group was fed with normal diet. The B group was fed with high - fat diet, the hot scale of fat, protein and carbohydrate was 59% , 21% , 20% , respectively. In C group the hot scale of fat, protein and Carbohydrate was 10% , 21% , 69%. We had glucose clamp technique and measured the related marker. The glucose clamp technique was operated on 5 rats which were selected from each group random. The process of glucose clamp technique was strictly in line with the reference. We geared the glucose infusion rate ( GIR) to maintain the stability of blood sugar, measuring the GIR in 60 - 120 minutes, un - operated rats draw the blood by cutting their tails, 2ml was used to measure TG, No and FFA, and 2ml was used to measure ET - 1 by way of anti - coagulate by EDTA and zymofren. The NO kit was provided by Nanjing Jiancheng Biology Engineering Institute, and the ET - 1 kit was provided by Beijing East - Asian Immune Reagent Institute. The thoracic aorta strip were used for observation specimen of electron microscope. All data were processed by SPSS, mono factor analysis of variance and linear correlation were used to analysis.ResultsThe weight of rats in B and C groups were higher than the ones of normal diet group, and the difference between them were significant.The operation group insulin, FFA and TG were higher than the ones of control group. The GIR of experiment group were lower than the ones of control group by the carbohydrate technique, and the difference between them were significant. NO decreased and ET - 1 increased, andthe difference between them and control group were significant. By the observation of electron microscope. The endothelial cells were abscis-sied and there were lots of incisura on nucleus, elastic lamina and the smooth muscle cells below it stretched to the cavum. The results of correlation analysis displayed that there was a negative correlation between GIR and insulin and weight, and no correlation with blood sugar. There was significant negative correlation between ISL and ET -1, FFA, and there was positive correlation with FFA and NO correlation with TG.Conclusions1. After feeding Wistar rats on high - fat and high - fructose diet, we can replicate IR rat model.2. There is significant alterations on NOET -1' level in the earlier period alteration in endothelial cells of IR rats.3. The significant increasing of FFA in rats feeding on high - fat and high - fructose diet can verify that the lipo - poision theory , and FFA is a major factor that lead to IR after birth.4.
