| To set up the gastroparetic models of diabetic rats, and observe the preventive and therapeutic effect of the complex prescription "Tang-An-Ye(TAY)" on DGP. To study its' possible mechanism so that it can be provided as a base of animal experiment for clinical application and further developing. Method:The diabetic rats were induced by injection with STZ(50mg/kg) into their abdominal cavities, then they were randomly divided into four groups: model group(â… ), injecting into stomach with TAY group(â…¡), TAL enema group(â…¢), Cisapride control group(â…£). Another 16 normal rats were chosen as the normal control group(â…¤) and fed with standard feedstuff regularly. All diabetic rats were fed on high calorie and fat feedstuff irregularly. During the experiment, the rats' common condition, such as appetite, the volume of urine and excrement, activity, weight, blood glucose were observed and noted. At the end of the experiment, all rats were injected with semi-solid paste into their stomaches. After 30 min, they were put to death, whose stomaches were taken out and weighed. Then the stomaches were weighed again after the contents being cleared up. So the rates of remained contents in the rats' stomaches were calculated. Then the circular smooth muscle segments were cut from the rats' antrums and their contractive ability was tested. The rest antrum was divided into two parts, one is for the research of immunohistochemistry, the other is for the observation by transmission electron microscope. Result:The rats'common conditions of â…¡,â…¢ were obviously better than that of model group, as well as better than that of â…£. The blood glucose, TG, CHOL and blood viscosity of â…¡, â…¢ were also better than that of model group.The rates of remained contents in the rats' stomaches in model group had a significant rise. Both the rates of â…¡and â…¢ were lower than model group. It could also be seen that the contractive frequency and amplitude of the circular smooth muscle of model group were lower than â…¤. The frequency and amplitude of â…¡ ,â…¢ were higher than that of the â… and â…£.The quantity of ICCs of model group had reduced obviously, the cells got small and most of their projections had disappeared. The close connection between ICC and nerve cells or smooth muscle cells reduced significantly. But the quantity of ICC of TAY treatment groups decreased slightly, most of them were normal, and the close connection still existed. By transmission electrom microscrope, it could also be found that the ICCs of model group had became small, cytoplasmic matrix had reduced, mitochondrions had decreased and varicosed, other organelles also decreased. The remained ICCs were rarely related with other cells. On the contrary, the ICCs' ultrastructures of TAY treatment groups were almost normal. Conclusions:The gastroparetic models of diabetic rats can be induced in a short time by injecting with STZ(50mg/kg) into their abdominal cavities and feeding them on high calorie and fat feedstuff. TAY can not only improve the common condition of diabeticrats, decrease the levels of blood glucose, TG, CHOL and blood viscosity, but also can improve the pathology change of ICCs obviously, enhance the contraction of gastric smooth muscle and promote the gastric empty. The TAY's enema has the same effect of improving the model rats' gastrointestinal activity as the method of injecting into stomach with TAY, so it is a safe and very useful therapic method to DGP. |
