Objective: To explore the pathogenesis of irritable bowel syndrome ( IBS ) and the mechanism of curing experimental rat model of IBS in Jiawei Tongxieyaofang. Methods: Model rats of IBS, which were prepared by chronic stimulation in colon, were randomly divided into four groups:the model group(Group B), the dicetel control group(GroupC),the low dosage of Jiawei Tongxieyaofang group (GroupD),the high dosage of Jiawei Tongxieyaofang group (Group E), with the addition of the normal control group (Group A). Different group received oral administration of different treatment in a same dose of O.lml/10gd for one month. And then ,to observe the bulgy capability of the sacculus that cause uplifting of abdomen and arching of back and the time of contract about abdomen muscle in rats.The level of 5-hydroxytryptamine (5-HT) in serum,substance P ( SP ) calcitonin gene related peptide ( CGRP ) in plasma were evaluated.Results: 1 After 7 weeks, the least bulgy capability of the sacculus that caused uplifting of abdomen and arching of back decreased markedly as compared with that in the normal group, and the time of contract of abdomen muscle increased obviously as compared with that in the normal group.lt showed model rats of IBS were establish.2 After 11 weeks , the least bulgy capability of the sacculus that caused uplifting of abdomen and arching of back and the time of contract of abdomen muscle all came back in three medicine groups.There was no obvious difference in three medicine groups.3 After 11 weeks ,the content of 5-HT in serum had a significantly increase in Group B as compared with that in Group A. In all three medicine groups, the content of 5-HT in serum all came back,and there was no obvious difference.4 After 11 weeks , the content of SP in plasma significantly increased in Group B as compared with that in Group A. Jiawei Tongxieyaofang obviously decreased the content of SP in plasma, while dicetel didn't.5 After 11 weeks , there was no obvious difference about the content of CGRP in plasma between Group B and Group A. The content of CGRP in plasma perhaps cound decrease in the model group.Only the high dosage of Jiawei Tongxieyaofang group increased the content of CGRP in plasma as compared with that in the model group. Conclusions: Jiawei Tongxieyaofang can get right the parameter about behavior and electrophysiology and adjust the level of 5-HT in serum and SP. CGRP in plasma in model rats. The synthetical effect of Jiawei Tongxieyaofang is better than that of dicetel,especially high dosage.
