Objective:1. To study the kinetogenic effects of 11 Chinese herbal medicines on colonic smooth muscle of rats in order to select the valuable one that could be studied further.2. To study the kinetogenic effects of Ah on colonic smooth muscle cells of rats and explore the contractile mechanism, in order to develop the new type of kinetogenic medicine on colon.Methods:1. The transverse and longitudinal contraction role of colonic smooth muscle strips of 11 Chinese herbal medicines and other medicines was separately observed.2. Smooth muscle cells were isolated from the colon of the rat. The contractile response of colonic smooth muscle cells to Ah and the effects of other medicines were separately observed.Results:1. The 11 Chinese herbal medicines can make traverse and longitudinal contraction at different levels (p<0.05).2. Ah can make dose dependent contraction with the concentration range from 1x10-6 mol/ L to 1 x10-3 mol/L, the concentration of the maximal response is 1 x 10-3 mol/L. Contraction caused by Ah could be inhibited by Atropin and EGTA, inhibition by Regidine and TMB-8 is not significant.Conclusion:1 .A screening study of the kinetogenic effects of 11 Chinese herbal medicines on the colon smooth muscle in rats : Semen Arecae, Fructus Foeniculi, Herba Taraxaci, Semen Raphani and Rhizoma Atractylodis Macrocephalae can make notable contraction on colonic smooth muscle strips of rats and worthy to be studied further.2.A study of the effects of Arecaline hydrobromide on the dispersed colonic smooth muscle cells in rats : The contraction caused by Ah is mediated by M receptor and depend on the presence of extracellular calcium.
