Objective:1. To determine six Chinese herbal medicines, such as Rhizoma Atractylodis Macrocephalae, Fructus Foeniculi, Herba Taraxaci, Semen Raphani ,Pericarpium Arecae and Rhizoma Curcumae whether has an effect on contractile activity of isolated guinea pig colonic muscle strips, and study their possible mechanism.2. To study the effects of Ah at different concentration on calcium mobilization in cultured smooth muscle cells of Rat.Methods:1. The colonic longitudinal muscle strips of guinea pigs were prepared.0.9% sodium chloride and acetylcholine were used as control. The effects of six Chinese herbal medicines and the control medicine on the contractibility of the colonic smooth muscle strips were observed. Contraction caused by Chinese herbal medicines could be inhibited by Atropine and Verapamil.2. The fluorescent intensity (FI) of intercellular calcium concentration ([Ca2+ ]i) was measured with confocal microscope in cultured smooth muscle cells of Rat loaded with Fluo-3/AM.Results:1.
