Stress is a widespread living fact that exsits, which all life need for survival anddevelopment, it is an important component part of the whole adaptive and protectingmechanism of life. However, the bad stress caused by mental and social factors, canlead to body obstacle and mental disorder more or less. Modern research approve,emotional stress has a influence that can not be ignored on human health, so it isimportant to study the animal model of stress and drug influencing stress directly. Thistask consist of two parts: theoretical study and experimental study ,discuss themechanism of stress and the effect of TCM.Theoretical study The following contents were mainly studied in this part: ①concept and stage ofstress; ② influence of stress on body; ③connection mechanism of stress withimmune ,neural and incretory system; ④the adjusting and controlling of stressreaction;⑤modern research progress on liver of TCM; ⑥relation of stress with liverof TCM.Experimental studyExperiment1 : Making and Evaluating The Rats Model of ChronicImmobilization Stress Sixty male rats were randomly divided into six groups and 10 rats in each group:the normal control group, model group of 7 days, model group of 21 days, the grouptreating with Sijunzitang or Xiaoyaosan or Jinkuishenqiwan( respectively simplifiedas the Sijunzitang group, the Xiaoyaosan group, the Jinkuishenqiwan group). Recipesused in the experiment were respectively from Formularies of the Bureau of People'sWelfare Pharmacies (Xiaoyaosan: Beichaihu 30g, Danggui 30g, Baishao 30g, Baizhu30g, Fuling 30g, Gancao fried with honey 15g, Shengjiang 10g, Bohe 10g;Sijunzitang: Dangshen 30g, Baizhu 15g, Fuling 15g, Gancao fried with honey 10g)and Synopsis of Prescriptions of the Golden Chamber (Jinkuishenqiwan: processedDihuang 60g, Shanyao 30g, Shanhzuyi 10g, Zexie 15g, Fuling 15g, Mudanpi 15g,Guizhi 10g, processed Fuzi 10g). Rats model of chronic immobilization stress wasmade by binding rats on the specific bound frame three hours a day for 3 weeks (1week in group B). Chinese herbs were made into powder by certain procedures forusages. Xiaoyaosan, Sijunzitang and Jinkuishenqiwan were respectively injected intothe stomach of rats in the groups D, E and F, the dosage are 5.82g, 2.62 g and 4.98gper kg,the cubage is 1ml per kg,the dosage is adjusted at any moment according to6 慢性束缚应激中枢 L-ENK 与肝主疏泄相关性研究the weight everyday of rats.The same cubage of distilled water was used for rats in thegroup A ,B ,C. Kill the rats of group F and get tissues after 1 week, deal with theothers 3 weeks later. Pay attention to observe appearance of rats during making model.Weigh the rats everyday and calculate the change of eating and drinking.Results: Common condition: the rats of model group appear listless, get together and donot like to move, do not resist actively, the hair of which have no luster, eat anddrink less and the stool of which is watery. The body weight and fodder intake: compared with rats of normal control group,fodder intake decrease evidently; Among groups treating with drugs, rats of the grouptreating with Sijunzitang eat the most. Weight gaining of rats of model groups andgroups of treating with drugs slow down compared with that of normal control group,the result is significant difference. Compared with that of model group of 21 days,Weight gaining of the groups of treating with drugs is obvious,but the result is notdifference statistically.Experiment 2: Ethology Study on Rats of Chronic Immobilization Stress The methods of grouping , making model and treating are the same asExperiment 1.
