Acute leukemia (AL) is clonal malignancy that is characterized by the appearance of increased number of immature myeloid cells in the marrow and blood. AL has been described as "xu lao", "re lao", "ji lao", "xue zheng" in Traditional Chinese Medicine(TCM). Chemotherapy is the main therapy of AL, and the drug of Chemotherapy can treat AL with inducing the apoptosis. Chinese herbal can relieve the poisonous and side effects, ehance the curetive effect, extend the period of remission, decrease relapse and there are many mechanisms of these effects, such as inducing apoptosis.We hold the opinion that the pathogenis of AL is asthenia combined with sthenia, dominated by heat toxin as well as blood stasis and sputum coagulation, insufficiency of the spleen and the kidney. Sophocarpidine can clear pestilence and counteract toxin, eliminate pathogenic heat from blood and stop bleeding, and promote blood circulation to dissipate blood stasis, that is to say, it can restrain leukemia cells.To explore the mechanism of Sophocarpidine in treating AML from inducing apoptosis and to supply the theorical basis in clinical therapy.Methods: 1. MTT assay detects the growth inhibition effect of Sophocarpidine on HL-60 cells.2. Morphological changes of apoptotic cells were investigated with Sophocarpidine on HL-60 cells.3. FCM assay detects the apoptotic cells and analysis cell cycle. Results: 1. MTT assay indicates: Sophocarpidine has obvious growthinhibition effect on HL-60 cells, and has dependency on concentration.IC50 is 0.2mg / ml.2. Invert microscope and fluorescence microscope indicates: There are some apoptotic body in HL-60 cells.3. Through the determination of FCM, the obvious apoptostic peak and S stage can be seen.Conclusion: Sophocarpidine can induce apoptosis in HL-60 cells. Inducing apoptosis is one of possible mechanisms in treating AL by Sophocarpidine.
