ObjectiveTo study the manufacture process and the quality standard of the WeiShu Granule in order to make up the shortage of the Traditional Chinese Medicine (TCM ) on decoction. To investigate the effects of the WeiShu Granule on the gastrointestinal motility and its function mechanism in order to use it more widely.To identify the toxicity of the WeiShu Granule in order to use it more safely.MethodThe manufacture process of the WeiShu Granule:firstly ,withdraw the volatile oil, which will be packaged by the 8-cyclodextrin;secondly, the medicine, which has been withdrew the volatile oil, will be extracted with water and precipitated with alcohol to obtain the concentrate; finally, the packaged volatile oil and the adapt dextrin will be added to the concentrate to make up of the WeiShu Granule.Use the thin-layer chromatography (TLC) to measure the concent of the chief herb in the WeiShu Granule, in order to evalute the manufacture process.The study of the function and the mechanism of WeiShu Granule:to observe the effect of WeiShu Granules on gastrointestinal motility in rat ,with the indicator-Dextran Blue-2000.To observe the Electrogastrography (EGG) of the gastric antrun, by placing an electrode in the gastric antrun and measuring the EGG by the BL-410 apparatus. To observe the content of blood plasma motlin (MTL), serum gastrin (GAS), the MTL and the GAS of the organization of the gastric antrun by the method of radioimmunoassay (RIA).The acute toxicity experiment of the WeiShu Granule: to observe whether the rats which are fed with the WeiShu Granule on first day will be dead in the following seven days. If there were rats dead, then anatomise the corpse and checks the important organs such as the liver, the stomach, the intestines, the heart, the lung by naked eye; and measure the dose of the WeiShu Granule which will lead the rats LD50.Else, observe the WeiShu Granule s maximum dose of one day.Result1 The experiment of the manufacture process indicates that all the index of the WeiShu Granule match the quality standard on granules in the Chinese medical dictionary (2000 version).The experiment of TLC indicates that the main compositions of each chief medicine all remains in the WeiShu Granule which is made by the above process.2 The experiment of gastrointestinal motility indicates that the WeiShu Granule can promote the gastrointestinal motility and antagonize the inhibition function of atropine, fenfluramine and dopamine on the gastrointestinal motility.3 The experiment of gastric antrun Electrogastro- graphy (EGG) indicates thatthe WeiShu Granule can enhance the swing and enhence the frequence of the EGG.4 The experiment of the change on hormone indicates that the WeiShu Granule can significantly increase the content of MTL, GAS in the blood and the the gastric antrun.5 The acute toxicity experiment shows that the LD50 cann t been measured. After given the maximum dose, which is 342 times of the healing dose, on the first day, there are no toxic reaction found and no dead rat observed in the followed seven days.Conclusion1 The WeiShu Granule which is made by the above process basically reserves the traditional herbs' attributes and absorbs the advantage of modern withdrawing craft at the same time.Compared with the soup, the WeiShu Granule is more scientific and more convenient.2 The WeiShu Granule can prompt the gastrointestinal motility, and the mechanism maybe includes the following: enhancing the function of the cholinergic nervous receptor and the 5-HT4, inhibiting the function of the receptor of D1 or D2, prompting the electrcity action of the gastric antrun, increasing the content of MTL and GAS in the blood and the gastric antrun.3 According to the acute toxicity experiment of WeiShu Granule, the medicine's toxicity is very low and its clinical application is more safe.
