By injected the whole 7721 cells as antigen into themouse abdominal cavity and picked out the spleen cells tofusing with myeloma cells, a new kind of monoclonalantidody Hepama-2 were screened. This kind of monoclonalantibody can specially combine with the cell-surfaceantigen on Hepatocellular Carcinoma cell line 7721. Weanalysed the characteristic of its antigen and theaffinity of this kind of monoclonal antibody. We alsoestimated the concentration of this kind of monoclonalantibody in the mouse ascites and batch-tissue culturesupernant. Othermore, we cloned its variable region geneand analysed the gene and protein sequence, prepared forthe next step of antibody humanization.
