Objetive To observe changes of expression of neurotrophin-3 (NT-3)the spinal neurons following complete spinal cord transection in rat , in order toprovide the experimental evidences for the mechanisms of regeneration andrepair after spinal cord injury and for neurotphins to treat spinal cord injury.Methods Adult Sprague-Dawley rats weighing 250g were divided into sixgroups at random: Normal group, sham operation group(n=8), and groups of theday 1,3,7,14 postoperative (n=8 for each time point). The spinal cord of ratwas transected compteted at the thoracic level 10. The expression of NT-3 inventral-dorsal horn of close together the spinal neurons was examined byimmunocytoche mistry anel imayt analysis. Results In normal group, theNT-3 positive cells were mainly localized in big spinal neurons and a little smallspinal neurons in ventral horn ,and in neurons in dorsal-horn. After operation,the expression of NT-3 in ventral-dorsal horn of the spinal neurons wasincreased and reached the peak at the day 1, there was a trend of decrease withtime in the groups of the day 3 and 7 ,and then returned to the normal level atthe day 14 in upside of the injury spinal cord ;after operation, the expression ofNT-3 in spinal neurons was increased at the day 1 and reached the peak at theday 3, there was a trend of decrease with time in the groups of the day 7, and thenreturned to the normal level at the day 14 in underside of the injury spinal cord .Conclusions The number of demand of NT-3 increases in the spinalneurons after spinal cord injury. The results suggest that NT-3 ptays an importantrole in the repair of spinal cord after injury.
