| Objective To evaluate the moblization of low, middle, high dosesAtorvastatin ,estrogen and Granulocyto-colony-Stimulating-Factor (G-CSF)to Endothelial Progenitor Cells(EPCs) of the rabbit, thus explore properdose ,medicine and time. Method 48 male New Zealand White rabbits wereaveragely divided into six groups :normal control , Atorvastatin 2.5mg ,Atorvastatin 5mg , Atorvastatin 10mg , estrogen and G-CSF group . EPCsnumber was surveyed at first, second ,third, forth week after administration.Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficolldensity gradient centrifugation , then the cells were plated on fibronectin-coatedculture bottles and dishes .After 3 days culture ,attached cells were collected.The number of EPCs was surveyed as PE-CD34 /FITC-CD133 dual-stainedpositive cells by Flow Cytometry Analysis . EPCs also could be characterizedby fluorescent microscope as cells double positive for FITC-UEA-1 andDil-acLDL .At third week , serum nitric oxide(NO) was measured andstatin groups had serum lipids scaled additionally .Result The curves ofmobilization of those medicines to EPCs varied as following : The nomalcontrol group had been low level base line, G-CSF group had kept higherlevel, Atorvastatin groups and estrogen group had shown sawtooth-like curves,which valleys were at the second week and peaks at the third week, EPCsnumber at the forth was lower than that at third week. Comparing with normalcontrol group , atorvastatin 5mg and G-CSF groups had significantly mobiliziedto EPCs(P<0.01) since the first week; atorvastatin 5mg group was highest at thethird week which was three fold to that of first week , and nearly 20 fold tonomal control group; atorvastatin 2.5mg group mobilizied EPCs at third andforth week ,P<0.05, P<0.01; 10mg group only at forth week P<0.05. Estrogengroup had similar effect to atorvastatin 2.5mg group thus stronger than it .Atforth week ,EPCs in five adminstated groups were higher than normal controlgroup .G-CSF group was highest ,other four groups among which there were nodifference was higher .Serum NO in atorvastatin 5mg and estrogen group werehigher, P<0.01, atorvastatin 10mg group was lower than normal controlgroup ,P<0.01.Serum lipids were similar among the six groups. ConclusionAtorvastatin ,estrogen and G-CSF groups could mobilize EPCs ,the groups andthe time points were dependent .The mobilization varied as following : G-CSF>atorvastatin 5mg >estrogen> atorvastatin 2.5mg >atorvastatin 10mg, and themobilition of statins and estrogen might relate to serum NO. The fuction ofatorvastatin groups had nothing to do with its modulation to serum lipids... |
