Objective:Ultrasound contrast agent of surfactant fluorocarbon-filledconsisted of Span-60 and Tween-80 was prepared. The relation of intensityof acoustic reflection in two-dimensions and dose of the agents investigatedin vitro. Ultrasound imaging of intensity of two-dimension and CDFI andPDI in hepatic portal vein and liver and kidney of normal rabbits wereobserved after intravenous injection of ultrasound contrast agent ofsurfactant . The increase in flow Doppler signal of liver VX2 tumor wasstudied after intravenous injection of surfactant ultrasound contrast agentS6T8 in vivo. Methods: San-60 and Tween-80 were mixed in a proportion of 1:1,then the mixed solution was sonicated at a frequpency of 20KHZ and asonication output of 110 watts, using a Misonix XL 2020 sonicator forthree minutes. The sonicated solution was stand for thirty minutes inroom temperature. The sonicated solution separates into three distinctlayers, with middle layer containing the desired products. The in vitroultrasound experiments were performed to assess acoustic backscatter ofthe agents and dose optimal needed to give the maximal the acousticenhancement. A respond dose of the agents was constructed. The powerDoppler flow signal was obtained in 5 rabbits with liver VX2 tumor.Intravenous injection of surfactant ultrasound contrast agent S6T8 wasadministered in a dose of 0.1mg/kg. The increase of intensity and time inpower Doppler flow signal was observed. Results:The diameter of the agents was found to distribute from 0.7to 8.0μm, with 90% of microbubbles less than 3.0μm. The mean size ofbubble was 1.81um and the bubble concentration was 7.2×109 /ml. Thehighest respond of the agents occurs at a dose of approximately 300μl.When the dosage was increased the intensity of acoustic reflection wasdecreased. An obvious increase of intensity in CDFI and PDI andtwo-dimension signal was observed in liver and kidney and hepatic portalvein of normal rabbit. An obvious increase of intensity in power Dopplersignal and two-dimension signal was observed in the surround and centreof liver tumor, and the period of enhancement of power Doppler signalsand signals of two-dimensions were long . Conclusions:The agents have the higher concentration and the bettersize distribution. The better intensity of acoustic reflection intwo-dimensions was obtained. Surfactant ultrasound contrast agent S6T8 iscapable of obvious enhancement of flow Doppler signals and intensity ofacoustic reflection in two-dimensions in normal rabbit liver and kidney byintravenous injections. It awaits further research.
