Orange peel is one of common Chinese herbs with long history in traditional Chinese medicine.Citrus plants are rich source of various physiologically especially"heath- promoting substance". It is commonly accepted that cancer formation can be prevented by the consumption of certain foods and flavonoids in Citrus are one of the most prominent cancer-preventing agents. In recent literature review on Citrus flavonoids, the polymethoxylated flavones found in the orange peels may be of even more interest. It is reported that polymethoxylated flavonoids such as nobiletin 3,5,6,7,8,3',4'- heptemthoxyflavone can inhibit the development of certain types of tumors and the proliferation of some cancer cell.This dissertation including nine parts is mainly about the effective fraction of polymethoxylated flavonoids from orange peels.Chapter 1 This chapter contains two parts. In the first part, documents about the research progresses in chemical constituents and pharmacological activities of Citrus plants in last twenty years were summed up and analyzed, 40 references were cited. In the second part, we made an overview about dropping pill made of Chinese herbs.Chapter 2 Five compounds were isolated from the effective fraction of orange peel. Three of them are polymethoxylated flavonoids.The orange peel extract was loaded onto a silica gel flash column and eluted by a mixed solvent system of ethyl acetate and cyclohexane, and the fractions collected. Upon combination of the eluted fractions, mainly containing nobiletin 3,5,6,7,8,3',4'- heptemthoxyflavone and natsudaidai.Chapter 3 The first part is to optimize the technology of extracting effective fraction of orange peel. The optimum technology of extraction was studied by orthogonal test using the content of nobiletin, 3,5,6,7,8,3',4'- heptemthoxyflavone and natsudaidai as selecting index. The factors of the orthogonal test table L9(34) were alcohol concentration(A), alcohol quantity(B), extracting time(C), and extracting times(D). As a result, the optimum parameters were A2B2C2D2 .The second part is to purify the alcoholic extract with liquid-liquid extraction and solid-liquid extraction. Through the purification, we get effective fraction of orange peel.
