Arteriosclerosis obliterans(ASO) is one of common chronic main and medium-sized artery ischemia disease, especially main medium-sized artery of lower limb's. It is the extremities' local manifestation of total Atherosclerosis(AS). It belongs to the "tuo ju" or "mai bi" range of TCM. There is always a puzzle about the etiology of ASO.The state of this illness is complicated and there are so many complications. It seriously hazardous to human's health because of it's high extremity amputation and fatality rate."WenMaiTong"is an effective formula to treat early ASO which includes Cassia twig, Membranous milkvetch root, Chinese angelica, Szechwan lovage rhizome. The role of it is warming Yang and boosting qi,activating the blood circulation and invigorating the meridian.Clinnical research indicate the formular could inhabit platelet adhesion reaction,prevent the thrombogenesis,down regulate fibrinogen, improve microcirculationand elastic of the artery.Animal experiment show that"WenMaiTong"has significant effect on experimental ASO of rabbit.It can improve the index of hemorrheology,repair the damage of the endothelial cell,decrese the density of ET in blood plasma,increase the density of NO in blood plasma.Modern medicine point out that the genesis of ASO was induced by muti-factors.The damage of theVEC is the prime factor and the adhension between monocytes and vessel wall was the committed step. This study applied many kinds of methods to evaluate the mechanism of"WenMaiTong"in preventing VEC from damage and decreasing the adhension between VEC and MC so as to provid academic and experimental support for clinical practice. Meanwhile, the science and rationality of this formula were proved by comparing the"WenMaiTong"whole formula with warming Yang and boosting qi part formula and quicken the blood and free the meridian part formula.In this study, HUVEC and THP-1 were taken as VEC and monocyte. The serum-pharmacological method was applied. The hyperlipemia serum was used as the induced factor. The cells were randomly divided into normal control group, hyperlipemia serum group,"WenMaiTong"high dosage group,"WenMaiTong"low dosage group, warming Yang and boosting qi group,quicken the blood and free the meridian group.
