Although western medicine has already made great progress in the studies on the pathogenesis and therapies for asthma, the incidence rate is still inclined to further increase. So it is very imperative to investigate the pathogenesis of asthma and find effective therapies. Traditional Chinese Medicine enjoys great history and inherent characteristics in treating asthma. Traditional Chinese materia medica and medicinal decoctions have multiple targets so that they can exert influences on multiple pathological processes of asthma and produce curative effects. With the help of modern biomedical sciences, we can demonstrate the therapeutic effects of Chinese materia medica and elucidate the underlying mechanisms of drug actions. These studies help us find new drugs for asthma from Chinese materia medica and medicinal decoctions. Jiawei-Sanao Decoction (MX) is prepared on the basis of ancient Sanao Decoction by Professor Qingguo Wang. The MX takes into consideration of main characters in pathogenesis of asthma including qi stagnation, phlegm abundance and blood coagulation. The decoction is very effective in controlling and preventing asthma attacks in clinical applications. The study mainly focuses on evaluating the antitussive and apophlegmatic effects, anti-asthmatic effects, broncho-relaxing effects and anti-inflammatory effects of MX. The objective of this study is to demonstrate the therapeutic effects of MX and partly elucidate their underlying mechanisms.1. ObjectiveDemonstrate the antitussive, apophlegmatic and anti-inflammatory actions of Jiawei Sanao Decoction (MX). Observe the anti-asthmatic effects of MX and the relaxation effects of MX on guinea pig tracheal smooth muscles ex vivo. Investigate the influence of MX on airway inflammation and the mechanisms underlying the anti-inflammatory action of MX.2. MethodsThe influence of MX on cough latent period and cough rate was investigated using strong aqua ammonia-induced cough model in mice to demonstrate MX's antitussive action. The effects of MX on the phenolsulfonphthalein excretion of mice trachea were observed to decide MX's apophlegmatic action. The anti-inflammatory effects of MX were detected by the degree of ear swelling using xylene-induced ear swelling in mice. The anti-asthmatic effects of MX were observed using OVA-induced asthmatic guinea pig model. In vitro experiment,guinea pig trachea strips were prepared. The bronchodilator effects of MX on isolated guinea pig trachea strips with or without the pre-contraction of bronchocontractiors (His and Mch) were studied. The response of OVA-induced isolated guinea pig trachea strips to OVA and the influence of MX on the response were tested.
