Objective The study intended to explore the main risk factors(including genetic, environmental, et al.) of coronary heart disease, the relationship of the genetic polymorphisms of PON2 and LPL and susceptibility to coronary heart disease, and the gene-gene or gene-environmental interaction ,so as to provide evidences for making the integrated preventive measures and revealing etiological facter's mechanisms for coronary heart disease.Methods A hospital-based case-control study which included 100 cases of coronary heart disease and 100 controls matched according to age,sex, collected from The fifth Hospital of Datong in Shan Xi province. All the objects were investigated in uniform questionnaires about risk factors for CHDs and were genotyped for PON2 C311S and LPL S447X polymorphism by polymerase chain reaction restriction fragment length polymorphism analysis (PCR-RFLP) method. All the data were analyzed by SPSS11.5 statistical software.Result (1) Comparing PON2 gene frequency between the two groups, it was showed that three genotypes were found in PON2C311S. The frequencies of CC, CS, SS were 23%, 11%, 66% in case group respectively, 54%, 38%, 8% in control group. There was significant difference between two groups at 0.01 level (χ2=72.818, P<0.001). Furthermore, with the increase of the number of allele S, the risk of CHDs was elevated. Allele frequencies of C and S were respectively 28.5%, 71.5% in case group, 73.0% and 27.0% in control group. The difference was remarkable between two groups.(χ2=79.228, P<0.001).(2)No significant difference was found in the frequencies of LPL S447X genotype and alleles between controls and CHD patients ( P > 0.05) . But when the subjects were divided into TC< 5.2mmol/ L group and TC≥5.2mmol/ L group ,or TG<1.8 mmol/ L group and TG≥1.8 mmol/ L group,or HDL≥0.9 mmol/ L group and HDL﹤0.9 mmol/ L group,or LDL﹤3.1 mmol/ L group and LDL≥3.1 mmol/ L group. the frequencies of SX/ XX genotype in the former were higher than that in the latter ( P <0.05) . So LPL SX/ XX genotype can do a beneficial influnce on serum lipoprotein profile.(3) By univariate logistic regression analysis,it was indicate that 17 factors (family histroy of CHDs,family histroy of hypertention, family histroy of diabetes, hypertention, diabetes, character of type A, working pression ,smoking, drinking, physical exercise , meal tasting, BMI, TG, TC, HDL, LDL, PON2C311S) were correlated with CHDs(P<0.05) in the levelα=0.05.(4) Analysis of multiple variables logistic regression showed that family histroy of CHDs, hypertention, diabetes , character of type A, , smoking, drinking, ,meal tasting, BMI , PON2C311S were the main risk factors of CHD, the value of OR were 4.887, 4.036, 2.578,...
