Whitmania pigra whitman has been used as a remedy for blood stasis and traumatic injury since hundreds and thousands years .Up to mow ,it is a main kind of medical herb used in Traditional Chinese Medicine for promoting blood circulation by removing blood stasis ,and often ,it is used after being processed .In modern medicine, Whitmania pigra whitman is used in the treatment of the thrombus .In this study the method of combining together chemical and pharmacological studies was employed in discovering the anticoagulant from Whitmania pigra whitman under the guidance of activity evaluation.The anticoagulant production response in vitro and in vivo was employed as the activity evaluating parameter to guide stepwise fraction evaluation.Activity evaluation showed that the activity of methanol extract from whitmania pigra whitman is stronger than the others. Skellysolve B fraction was the abstracted with methanol,acetic ether,n-butanol and water in turn. Acetic ether fraction showed stronger activity than the others.The chemical constituents of Skellysolve B fraction were analyzed by GC-MS. 16 components were identified. The major components are Octadecanoic acid , Tetradecanoic acid methyl ester1, Heptadecanoic acid, 14methyl-pentadecan oic acid,methyl ester , 10-Octadecenoic acid methyl ester,ctadecanoic acid methyl ester, 10-nonadecenoic acid methyl ester,13-docosenoic acid methyl ester and etc.A compound was isolated from an acetic ether fraction and identified as cholesterol.
