Chemical Constituents of Whitmania pigraHirudo, as a representative animal medicine for promoting blood circulation and removing blood stasis, has been used for more than two thousand years. It is the dry body of Whitmania pigra Whitman,Hirudo nipponica Whitman or Whitmania acranulata Whitman. Advances in the research of chemical constituents and pharmacological activities of leech were reviewed in this thesis, which provides the basis of rational development and utilization of resource for this animal drug.In addition, for revealing the drug action material, drug usage and resource foundation. Chemical constituents of Whitmania pigra were investigated.9 compounds were isolated from part of its dry body extracts by means of the modern chromatographic techniques.3 structures have been confirmed by the physicochemical properties of the chemical compounds and the spectroscopic data and identified as:Cholesterol, Cetyl ethers of glycerol and campesterol, the last two were isolated from Whitman animals and this animal for the first time, respsctively. Extraction process of Shenyuan TabletShenyuan Tablet is the new traditional chinese medicine which cures coronary heart disease and angina pectoris. It consists of Panax quinquefolius.L. and Corydalis yanhusuo W. T. Wang, which is used to soften the blood vessel, eliminate stasis, prevent and cure coronary heart disease.Three different groups of Panax quinquefolius.L. and Corydalis yanhusuo W. T. Wang of Shenyuan Tablet were qualitatively identified by thin-layer chromatography (TLC) and determained according to Chinese Pharmacopoeia.With the content of ginsenoside Rg1,ginsenoside Re, ginsenoside Rb1 and tetrahydropalmatine as evaluation indexes, first through single-factor test to determine the extraction solvent and finally choose the best solvent is 70% alcohol, then choose L9(34) orthogonal experimental arrangement, inspect the solvent amount(A), extraction time(B) and times(C) as three factors to optimize the process, each factor has three levels. HPLC method is first used to determine the ginsenoside and tetrahydropalmatine content of Shenyuan Tablet, which provides a preliminary scientific basis for the quality control of the Tablet. By single-factor and orthogonal test, the result of the best extraction condition is extracted with 8 times amount of 70% alcohol,2 hours for 2 times. Experiment results verify the extraction process and prove its rationality and feasibility, it also provides a reference for large-scale production. HPLC analysis is also first used to determine the content of Shenyuan Tablet. The results show that the determination method of Shenyuan Tablet is convenient, the extraction process is reasonable, feasible and quality control.
