Background: Many transcription factors have now been discovered and several of them are involved in asthmatic inflammation, including nuclear factor- κ B (NF-κB), activator protein-1(AP-1) and cyclic AMP response element binding protein (CREB). The gene ESE-3 codes for the protein that belong to an ETS transcription factor subfamily and its expression is restricted to primarily glandular epithelium. ESE-3 was expressed constitutively in human bronchial epithelial cells (HBEC) and mucous gland epithelial cells, however, ESE-3 expression was induced in human bronchial smooth muscle cells (HBSMC) and fibroblasts (HBFB) after treatment with inflammatory factor related to asthma. Glucocorticoid inhalation is an effective therapy generally accepted for the treatment of bronchial asthma. However the exact roles of ESE-3 in atopic asthma and the influence of glucocorticoid inhalation are still largely unknown.Objective: To investigate the expression of ESE-3 mouse homology gene Ehf in asthmatic mouse and to explore the role of ESE-3 in human bronchial asthma. To survey the influence of glucocorticoid inhalation on ESE-3 in addition.Methods: Thirty female mice (BALB/c) were randomly divided into three groups : control group, asthmatic model group and treatment group(treated with budenide). Expression of Ehf mRNA was detected by the methods of reverse transcriptase-polymerase chain reaction (RT-PCR) and Real-Time PCR, Expression of protein was detected by the methods of Western-blotting. Results: Ehf was expressed in both control group, asthmatic model group and treatment group, Expression level of Ehf mRNA in asthmatic model group was 0.5800 ±0.2081, significantly higher than that (0.2300 ± 0.2336) in control group(P<0.05) and that (0.2930 ± 0.2117)in treatment group(P<0.05) by the...
