Objective: we have observed there are nonrandom losses of chromosome segment in mass human tumors due to deep study on tumor chromosomes and genes ,with the development of tumor molecular biology and gene engineering. The result indicates that the region of these chromosome segment may contain genes associated with the origin and progression of human tumor .In near passed years , we found that there are tumor suppressor genes (TSG) close to a loci on chromosome ,if there was highly frequent LOH (loss of heterozygosity) on the loci through screening special PMM (polymorphic microsatellite marker) on genes of tumor tissues and the corresponding normal tissues. For many tumor suppressor genes were discoverded by studying LOH , in vivo ,analysis of LOH becomes one of the important methods screening inactivity of tumor suppressor genes ,even discovering or locating new ones.AT present chromosome 12p is a hot region .Highly frequent LOH on chromosome 12p in many tumors such as lung carcinomas, FL and ALL. indicates the region may contain one or many tumor suppressor genes. But in vivo there are no reports about the study of LOH on chromosome 12p in the world. Here, four polymorphic microsatellite markers on chromosome 12p in 28 cases of gliomas were analyzed to detect LOH by PCR based techniques. The study was performed to locate the deletion areas probably harboring tumor suppressor genes and provide clues for discovering tumor suppressor genes.
