Parkinson's disease (PD) is the second most frequent neurodegenerative disorder. It is characterized neuropathologically by progressive loss of dopaminergic neurons in the substantia nigra.The causes of PD are unknown but considerable evidence suggests a multifactorial etiology involving genetic and environmental factors. In recently study toxin-induced mitochondrial disfunction has been suggested to play an important role in the pathogenesis of PD. Paraquat (PQ) is familiar to MPP~+(1-methyl-4-phenylpyridinium), which is a classical tool in making PD model. PQ is also a world-wide-used herbicide in the past decades. There are more and more evidence came from dependability study on PQ and PD that PQ may induce PD.Pheochromocytoma cell line (PC12) is the frequent used dopaminergic cell line in making PD cell-model. Study on the mechanisms involved in toxininduced PC12 degeneration by PQ could reveal new insights concerning possible neuroprotective treatment of PD or PQ injury.In the first part we investigated PQ-induced neurotoxicity in PC12 cells and the possible mechanisms. We found that PQ caused cell damage time- and concentration-dependently. PQ-induced-cell death had the characterization of apoptosis in these tests, it was likely increasing the ecto-cellular levels of nitric oxide (NO) and decreasing the levels of total antioxide capability in a concentration-dependently way. NO-induced oxidative stress plays an important role in PQ-induced neurotoxicity in dopaminergic cells by strengthening T-AOC of injuried neuron.In the second part we studied the possible protective effect on PQ-injury of some effective drugs used in clinical treatment. We found that Amantadine, Benadryl, Dopamine, Ascorbic acid reduced PQ-induced neurotoxicity of PC12 cells markedly. Taken together, these data demonstrate that treatment with PQ may induce acuteoxidative neurotoxicity in PC 12 cells —— such a phenomenon is familiar to PD,moreover, such an injury could be resisted with some PD medication.
