Objective: To discuss the effect on Radix Polygoni Multiflori lowering STZ rats’ blood glucose and explore its possible mechanism.Methods:48 SD rats were randomly divided into control group and experimental group, first, the rats of experimental group were induced in type 1 diabetic with high-dose streptozotocin by intraperitoneal injection for 3 weeks, the model was built successful when those rats’ blood glucose level≥16.7mmol.L-1 lasting 3weeks. then type 1 diabetic rats were randomly divided into type 1 diabetes model group, Low-dose group of Radix Polygoni Multiflori, High-dose group of Radix Polygoni Multiflori; The control group were gavaged citrate sodium citrate buffer first 3 weeks, later 8 weeks,they were given normal saline; the rats of type 1 diabetes group were given normal saline; The Low-dose group of Radix Polygoni Multiflori at the dose of 1g·kg-1·d-1Radix Polygoni Multiflori,The High-dose group of Radix Polygoni Multiflori at the dose of 2g·kg-1·d-1Radix Polygoni Multiflori. until the 12 th week. the rats’ body weight, blood glucose levels were measured in the first five weeks and 12 weeks,and the rats’ amount of water, food intake and other general conditions were observed; To get pancreas tissue after anesthetizing rats and make tissue sections through the process of being fixed、embeded;To make HE staining, to make immuno histochemical method detecting β cell insulin secretion and calculate the percentage of insulin secretion positive cells in 100 islet cells, to marke the apoptosis of STZ rats’ pancreas by TUNEL and calculate the apoptosis rate among 100 ialet cells. Results: At fifth week, compared with the control group, the weight of rats in each experimental group were significantly decreased(P<0.05); blood glucose was significantly increased(P<0.05). Compared with type 1 diabetes mellitus rats, Low-dose group of Radix Polygoni Multiflori’s weight increased not obviously(P> 0.05), their blood glucose were lowered not obviously(P> 0.05); High-dose group of Radix Polygoni Multiflori’s weight were increased obviously(P<0.05), their blood glucose were lowered obviously(P <0.05);while at 12 nd weeks, compared with type 1 diabete’s group, Low-dose group of Radix Polygoni Multiflori rats’ weight were increased not obviously(P> 0.05);and blood glucose lowering not significantly(P> 0.05),the Symptoms of diabets’ rats were not obviously improved. islet Cell apoptosis index decreased not significantly(P > 0.05); insulin-positive expression was not improved Significantly(P> 0.05).Compared with type 1 diabetes group, High-dose group of Radix Polygoni Multiflori rats’ weight were increased obviously(P< 0.05);and blood glucose was lowered significantly(P<0.05),the symptoms of diabets’ rats were obviously improved. islet cell apoptosis index decreased significantly(P < 0.05); insulin-positive expression was improved significantly(P<0.05). Compared with the Low-dose group of Radix Polygoni Multiflori, High-dose group of Radix Polygoni Multiflori were generally better;weight was increasd significantly(P<0.05),blood glucose was significantly lower(P<0.05),the symptoms of diabets’ rats were obviously improved. islet cell apoptosis index decreased significantly( P < 0.05); insulin-positive expression was improved significantly(P<0.05). However, compared with the control group,the High-dose group of Radix Polygoni Multiflori rats’ diabetic symptoms has some improvements, but still did not reach the level of a complete cure. Conclusion: 1, Radix Polygoni Multiflori can lower the diabetic rats’ blood glucose,and the result has a positive effect with the increasing of Radix Polygoni Multiflori.2, Radix Polygoni Multiflori can reduce the STZ rats’ blood glucose and protect the islet cell from Progressive damaging,which may be involved to the inhibition of islet cell apoptosis and improve the expression of insulin.
