Objective To investigate the relationship of Transforming growth factor beta 1 (TGF—β1)gene T869C polymorphism with type 2 diabetes mellitus and diabetic nephropathy(DN). Methods The polymorphisms of TGF β1 gene T869C(Leu10Pro) was analyzed by the sequence specific primers polymerase chain reaction(PCR-SSP) methods in 100 patients with type 2 diabetes mellitus, including 44 cases without albuminuria(NDN group), 56 cases with micro- or macroalbuminuria(DN group) and 47 healthy controls. The distribution frequencies of the genotype and allele as well as relative clinical date were compared among different groups. Results (1) The genotype distribution of the TGF—β1 gene T869C polymorphism was significant differences among 3 groups(X~2=11.158, P=0.025), and the allele frequency also was significant differences (X~2=10.215, P=0.006). (2) The type 2 diabetes mellitus group has higher frequency of CC/CT genotypes than the control group (43.0% VS 23.4%, X~2=5.283,P=0.022) . C allele frequency is also higher in the DM group than NC group (64% VS 47.9%, X~2=6.868, P=0.009) . The relative risk suffered from type 2 diabetes mellitus of C allele carrier was 1.94 times of the T allele carrier(OR=1.936,95%Cl 1.178—3.182).
