Objective: Acute cholangitis of severe type(ACST)is acute inflammation for bacterial infecting biliary system. If to move further, multiple organ dysfunction will happen. We selected two genes macrophage migration inhibitory(MIF) and toll-like receptor 4 (TLR4) and monitored change of their expressive quantitative in liver ,lung and hear of ACST rat model.To study associativity of MIF ,TLR4 and Tumor necrosis factor (TNF),this experiment can offer new research route and data about pathogenesis of ACST originating multiple organ dysfunction.Method: The expression of MIF and TLR4 at 2h,6h,12h,24 and 36h in three kinds of tissue are monitored by Real Time PCR. By radio-immunization we determin content of TNF in blood of ACST rat model and make time change curve, pathology changes in form are observed by HE drum.Results: 1. It has been showed that degree of injury of liver aggravates gradually following change of time. Pathology changes of lung and heart are no obvious.2. It has been showed that expression of the two genes (MIF,TLR4)in three kinds of tissue of ACST rat model has coherent time regular pattern ,and is higher than in the control group.3. It has been showed that content of TNF in blood higher than in the control group, and no prominent difference at 2h,6h,12h,24 and 36h.Conclusion: It has been showed that there are higher expression about MIF and TLR4 in the ACST rat model tissue than in the control group...
