| Shuang Huang lian for injection is a kind of traditional Chinese medicine powder forintravenous injection, which is from three species of Chinese crude plant: Lonicera japonica, Scutellaria baicalensis Georgi, and forsythia suspense vahl after a series ofprotocols of extracting, purifying, spray drying(or freeze drying) and sterilized packing. It possessed notable anti-bacterial, anti-viral, immunostimulant activity. In this thesis, the CPE method was used to test the anti-virus activity of every fraction., the constituents of active parts were studied, which make good foundation for the establishment of finger print spectrum of Shuang Huang Lian for injection. The result showed that the parts (including EtOAc Fr., n-BuOH fr. and water fr) abstracted from Shuang Huang Lian for injection exhibited anti-virus activity. Fourteen compounds were isolated from EtOAc Fr. and n-BuOH fr. , and twelve of them were determined on the basis of physico-chemical properties and spectral data as galactitol(1), p-hydroxy benzeneaceticacid(2), baicalein(3), wogonin(4), luteoloside(5), isolariciresinol(6), baicalin(7), rengynol(8), chlorogenic acid(9), daucosterol(10), β-sitosterol(11), phillyrin(12).On the basis of chemical constituents study, fingerprint spectra of Shuang Huang Lian for injection were also studied. To establish the fingerprint spectra of Shuang Huang Lian, we adopted chlorogenic acid, baicalin and phillyrin as references. The HPLC system consists of an ODS column, detection wavelength at 350nm, flow rate was lml/min, heated at 30℃, the mobile phase was 0.25% acetic:methnol for gradient elution in HPLC. We apply the similarity degree evaluating system of traditional Chinese medicine finger print spectrum for the chromatogram of samples evaluated. By comparing the fingerprint spectra with that of the control, calculating the similarity degree, the result was worked out. With the method above-mentioned, we have mensurated the fingerprint spectra of medicinal materials, intermediate, sample of reference and finished products. The results indicated that, there were good pertinency between medicinal materials, intermediate and finished products. The result accorded with the claim on the fingerprint spectra. |
