Radix Codonopsis is the roots of Codonopsis pilosula (Franch.)Nannf.Codonopsis pilosula Nannf. var. modesta (Nannf.) L. T. Shen.and Codonopsistangshen Oily. It's a common Chinese herb with long history in traditional Chinesemedicine.We study the standardization of processing technology and the standard forquality control.This dissertation including six parts.Chapter 1 A review of the literature studies on six aspects including theprocessing history evolution,the existing processing technique, the chemicalconstituents,the pharmacological effect,the research survey of 5-HMF, the briefsummary and argumentation.87 references were cited.Chapter 2 To study the chemical constituents of Radix Codonopsis. The extractfrom the roots of Radix Codonopsis was subjected to chromatography on silica geland Sephadex LH-20 column.The compounds obtained were identified on the basisof their physicochemical and spectroscopic evidences. Six compounds wereisolated and their structures were elucidated as taraxerylacetate,friedelin,△~7-stigrnasta-7-ene-3-one,Stigmasta-5,22-diene-3-one,taraxerol,and5-hydroxymethyl-2-furaldehyde.Chapter 3 Establish methods of determining the special compounds in RadixCodonopsisFirstly,To establish the UV method for content determination of totalpolyacetylenes in Radix Codonopsis.using lobetyolin as reference substance Thecalibration curve is linear at ranges of 5.28~26.4ug/ml(r=9997). The averagerecoveries were 99.01%(RSD=2.96%,n=5).Secondly,To establish the HPLC method for content determination of lobe-tyolin in Radix Codonopsis.using lobetyolin as reference substance. The calibrationcurves are linear at ranges of 0.264~1.32ug, The average recoveries were95.96%(RSD=0.70%,n=5).Thirdly, To establish the HPLC method for content determination of 5-HMFin Radix Codonopsis. using 5-HMF as reference substance. The calibration curves arelinear at ranges of 0.0864~0.2016ug The average recoveries were99.85 %(RSD=2.92%,n=5).Then To establish the VIS method for content determination of totalpolysaccharose in Radix Codonopsis. using D-glucose as reference substance. The calibration curve is linear at ranges of 14.4~86.4ug/ml,The average recoverieswere 98.10% (RSD=1.64%,n=6).Chapter 4 discusses the studies on the processing technology. It is divided into 3parts.Part 1 Discusses the studies on the cutting technology of Herba RadixCodonopsisTo optimize the cutting technology of Herba Radix Codonopsis withorthogonal design,We select the immersion time,moist time and dryness manner asreview factors and use ethanol extraction to optimize the technology.The optimumcutting condition obtained was: immersion 15 mins,moist 3 hours, drying within 50℃.Studies on the cutting technology in factory of Herba Radix Codonopsis.Afterthe factory manufacturing, the cutting technology in factory with clear technologicalparameters which can be easily controlled was established.The cutting method ofHerba Radix Codonopsis was as follows: get rid of fragments and other weeds fromthe whole Herba Radix Codonopsis, immers 15~20 mins,use enough warter,moist3 hours, cutting thick slices or small cubes,drying within 50℃.Part 2 To study the processing principle of fryed Radix Codonopsis UseHPLC method comparising pringtings with Radix Codonopsis and it's processedproduct. Isolate and purificate processed products. Elucidate the specialcompound's structure by determining it's physical-chemical properties and spectraldata.In order to approach the compond's generation pathway.Drying oven heatingmethod was used to simulate process experiment.As a result, Polysaccharides andorganic acid such as Ferulic acid reacting in high temerature is the main pathway togenerate 5-HMF, monosaccharide in weak sour or high temperature conditions canreact by itself to generate 5-HMEPart 3 Discusses the studies on the firing technology of Herba RadixCodonopsis.Analysing many factors such as property.Manipulation.stuff to choice a methoed forfiring.It is firing Radix Codonopsis cubes with bran.Then discusses the technologicalparameters. We select the amout of bran, firing temperature and firing time as reviewfactors and use the content of 5-HMF as the symbol constituents.The optimum firingcondition obtained was: Compare with the amout of Radix Codonopsis cubes using50% bran, 150℃,firing 5 mins.Studies on the firing technology in factory of Herba Radix Codonopsis.Afterthe factory manufacturing, the firing technology in factory with clear technological parameters which can be easily controlled was established.The firing method ofHerba Radix Codonopsis was as follows: Use Radix Codonopsis cubes, the amourof bran is 50%,in low temperature put cubes and bran into firing machine,keep theinside temperature maintaining 170~230℃, stir-fire 2~3min, 40r/min.Chapter 5 Discusses the studies on the standard for quality control of Herba RadixCodonopsis.The studies on the property, micro-identification,chemical identifica-tion. Identifications by TLC method of Herba Radix Codonopsis and processedproducts were performed in this thesis for the lack of these items in ChinesePharmacopoeia (2005 edition). Moreover, the water, total ash,extractivesremained.The quality standard of different decoctions were drafted on the base ofanalyzing 10 batches samples.The quality standard mainly include:property,micro-identification,chemical identification,identifications by TLC method,examination for impurities,contents of water, total ash, extractives, total lobetyolin or5-HMF.Chapter 6 analysis and summary...
