Agreat number of studies confirm that the insulin resistance(IR) is an importantfactor of diabetes and the research of insulin resistance has become hotspot of themedical science studies nowadays. Evaluation method of insulin resistance includeseuglycemic hyperinsulinemic glucose clamp Technique (GC),steady state model, countdown of FPG and FINS product, etc. Among them, Euglycemic Hyperinsulinemic Glucoseclamp technique is accepted generally in the nations as "golden standard". It hasalready a lot of methods for insulin resistance animal model establishment, such asinherited model, the food induced model, the medicine induced model, model inducedby food with small quantity of the medicine and surgical operation, etc. The modelinduce by food or induced by the food with small quantity of the medicine is closewith the natural course of illness of insulin resistance, economic and stable, andit is considered to be an ideal animal model of insulin resistance now. We successfullybuilt up the model in Fructose-Fed Rats (FFR) and we applied the EuglycemicHyperinsulinemic Glucose clamp technique to evaluate the model. We usedinternationally accepted method of estimating insulin resistance and FFR model toprobe into ameliorative effect of compound herbs Tang Shen capsule on insulinresistance and some mechanisms. This research aimed at building up fat rat model byhigh fat feeding and observing the role of Tang Shen capsule in the improvement ofinsulin resistance and blood lipid. The purpose of the study, therefore, is to buildup the insulin resistance obese model induced by high fat feeding and observe theeffect of Tang Shen capsule on insulin resistance and blood lipids of obese rats withinsulin resistance.Six-week-old male Sprague-Dawley rats were divided into chow group (60%vegetable strach, 11% fat and 29% protein) and high fat feeding group (78.8%basefeed, 1%Cholesterol, 10%yolk, 10% lard, 0.2%Bile Salt). After 8 weeks, the highfat feeding group is further divided into 3 sub-groups again, they are model group,Tang Shen capsule group, and AVANDIA group. The rats in model group were fed withhigh lipid, those in Tang Shen capsule group were fed with Tang Shen capsule(60mg/kg/day), and those in AVANDIA group were fed AVANDIA (2mg/kg/day). The medicaldrugs were dissolved in physiologic saline, and the rats in chow group and the modelgroupwere fed physiologic saline at the equal content. After 4 weeks of the experiment,test with the guglycemic Hyperinsulinemic Glucose Clamp technique was done. Theindexes were composed of fat wet weight, M value, weight, systolic blood pressure,TG, TC, LDL-C and HDL-C.Results showed that viscera fat wet weight and avoirdupois of the Sprague-Dawleyrats fed by high fat feeding (model group) were significantly increased compared to those of chow group. Insulin sensitivity (M value) significantly reduced in modelgroup compared to that of chow group. The rat model is characterized by typicalinsulin resistance and fatness. The Tang Shen capsule improved significantly M valueof the Sprague-Dawley rats, exhibited a decreasing trend of viscera fat wet weightand TC values.In cluonclusion, the Tang Shen capsule can improve significantly insulin resistanceand blood lipid metabolism.
