AIM:To explore the effects on differentiation to neurons and astricyte,of neural stem cells (NSCs )after electric stimulation of cerebellar fastigial nucleus and the influence of coexistence on differentiation to nerons and astricyte from its.MOTHODS:128 same genetic background rats were randomly divided into 4groups: NSCs group, (fastigial nucleus stimulation) FNS and NSCs group, coexistence body group, FNS and coexistence body group. This experiment we used neonatal same gene rat to get cells .The coexistence body was composed of NSCs isolated from hippocampi , vascular endothelial cells isolated from cerebra cortex and laminin. The rats were stimulated the cerebellar fastigial nucleus 1 hour before MCAO operation ,24 hours after MCAO operation ,we would separatedly transplant NSCs and coexistence body to the ischemic penumbra of MCAO rats. The rats from NSCs group coexistence body group had not been stimulated the cerebellar fastigial nucleus .The NSCs were labeled with Brdu before transplantiation .we would observe differentiation of NSCs throught immunofluorescence staining at 3days,7days,14days,60days after transplantiation.RESULTS:At 7day,14day,60day,the differentiation percent of FNS and coexistence body group was higher than that of FNS and NSCs group , the differentiation percent of FNS and NSCs group.is higher than that of NSCs group.there was difference between each group (all P<0.05).CONCLUSIONS:Electric stimulation of cerebellar fastigiul nuclus and coexistence body were both improved differentiation to nerons and astricyte of neural stem cells transplanted to MCAO rats, coexistence body and electric stimulation of cerebellar fastigial nucleus improve the differentiation to nerons from neural stem cells in coordination.
