Objective To establish stabile neurogliocytoma cells vitro primary culture model and carry out vitro chemosensitivity experiment by the MTT assay. To provide theory guidance for designning and optimizing clinical chemotherapy scheme, to practice individualization chemotherapy.Method using 32 fresh specimen which were proved to be neurogliocytoma to produce monoplast suspension and carry out vitro primary culture, add 5 antineoplastic drugs with peak plasma concentration in it, after 72 hours measure the survival tumor cell with the MTT colorimetric assay, get the optical density and the inhibition ratio, then judge the sensitiveness and drug fast of every drugs to every tumors, to guide the clinical individual Chemotherapy.Result succeed in primary culture and susceptibility test of 32 clinical tumors specimen. There are 14 times that the inhibition ratio exceeds 50% in the test of DDP with peak plasma concentration;3 of VCR;20 of VM-26;9 of VP-16;7 of paclitaxe. The order of sensitivity is: VM-26>DDP>VP-16> paclitaxe >VCR. There are 29 tumors specimen sensitive to one drug or more of 5 drugs, 21 tumors specimen sensitive to two drugs or more of 5 drugs.Conclude MTT colorimetric assay is convenient objective simple method with good repeatability. Every one of 5 drugs commonly used has the situation of drug resistance. Chemosensitivity assay can avoid using the resistant drugs and provide guidance to improve the clinical chemotherapy effect.
