Rhizoma Curcumae was used in traditional Chinese medicine for the treatment of various cancers such as cervical carcinoma, vulva cancer, skin neoplasm, thyroid tumor, esophageal neoplasm, gastric and intestinal cancer. And its essential oil functioned as an anti-tumor, anti-virus, anti-inflammation, anti-early pregnancy and anti-thrombus agent in pharmacological studies and was widely used as a clinical remedy for virus and cancer. Curcumol is one major constituent of Curcuma aromatica oil. It has been reported that curcumol also displayed anti-liver fibrosis in recent years. Curcumol, which chemical name is: 6H-3a, 6-epoxyazulen-6-ol, octahydro-3-methyl-8-methylene-5-(1-methylethyl)-(3S, 3aS, 5S, 6R, 8aS), is one of the major active components of the essential oil with the structure of sesquiterpene hemiketal.Three microorganisms (Aspergillus carbonarius IFFI 02087; Syncephalastrum racemosum AS 3.264 and Penicillium urticae IFFI 04015) were employed to bio-transform the substrateā€”curcumol, ten products were isolated and purified. The structures of 8 of them were determined on the basis of chemical and spectroscopic data. They are characterized as: 2a-hydroxy curcumol (Product CA-1, CS-5), 15-hydroxy curcumol (Product CA-2), 3a-hydroxy curcumol (Product CA-3, CP-8), 4a-hydroxy curcumol (Product CS-4), 14-hydroxy-9-en-curcumol (Product CS-6), 11 R-12-hydroxy curcumol (Product CP-7), respectively. Among of them, Product CS-4 is a new compound. And product CA-1 or CS-5 was also isolated from the broth of both microorganisms, which has been isolated from the urine of rats.This study would provide more curcumol derivatives for screening pharmacologically activities. On the other hand, product CA-1 or CS-5 was both isolated from the broth of microorganisms and the urine of curcumol-dosed rats, which showed the metabolic similarity of curcumol in between metabolic model of fungus and mammals.The method of Two-step fermentation was used to study the cultured-condition of Aspergillus niger. The optimized seeds medium was obtained. And the transformation efficiency of a single compound was evidently raised.
