Objective To investigate the effects of combined use of Valsartan and Benazepril on NF-κB in glomeruli of diabetic rats,and study their possible mechanisms.Methods We divided the experimental rats into five groups:non-diabetic controlrats(NC); STZ-induced diabetic rats(DM);diabetic rats treated with Valsartan(V);diabetic rats treated with Benazepril(B);diabetic rats treated with Valsartan and Benazepril(V+B).After 8 weeks, creatinine clear rate(Ccr),urinary albumin excretion(UAE)were measured.Expressions of NF-κB were detected by immuno-histochemical staining.ResultsCcr,UAE,expressions of NF-κB in diabetic rats and all treated groups were significantly higher than non-diabetic controlrats;Ccr,UAE,expressions of NF-κB in diabetic rats treated were significantly lower than untreated rats,.There were great depression in UAE,expressions of NF-κB between V+B group and V or B group.The relationship of NF-κB was positively correlated with urinary albumin excretion and creatinine clear rate.Conclusion Valsartan and Benazepril has renal protective effect partly through reducing NF-κB expression.Combination of both offer further benefits,arrest proteinuria and protect from renal function and structure impairment.
