Objective:To study the protective effects of apigenin in myocardium of rats with ischemia reperfusion(I-R)injury and investigate the cardioprotective action problem mechanism.Methods:Myocardial ischemia reperfusion injury model was established through occluding the left anterior descending branch of coronary artery for 10min and removing the ligation later to reperfuse for 10min in vivo.Different doses of apigenin were administered through intravenous injection 5 min before the onset of ischemia.We observed the antiarrhythmia effects of apigenin during reperfusion.After reperfusion,we measured myocardial injury sizes and myocardial infarct sizes(MIS)by dying injury ventricles of I-R rat in TTC solution,measured the level of myocardial superoxide dismutase(SOD),myocardial malondialdehyde(MDA)and serum lactic dehydrogenase(LDH)through UV—Spectromete.We studied the protective effects of apigenin in myocardium of rats with ischemia reperfusion(I-R)injury and explored its action mechanism.Results:As compared with the ischemia reperfusion,1,Apigenin(4 mg/kg)could suppress the incidence of ventricular tachycardia(VT),ventricular fibrillation(VF)and arrhythmias,decrea -se arrhythmia scores,shorten the duration ofarrhythmia(P<0.05).2,Apigenin(1mg/kg,2 mg/kg,4mg/kg)could reduced myocardial MIS in a dose-dependent manner(P<0.01).3,Api -genin(2mg/kg,4mg/kg)signifisantly improved myocardial SOD activity(P<0.05).4,Api -genin(2mg/kg,4mg/kg)remarkably decreased the content of myocardial MDA(P<0.01).5,Apigenin(1mg/kg,2mg/kg,4mg/kg)could significantly reduce the level of serum LDH in a dose-dependent manner(P<0.01).Conclusion:Apigenin can prevent myocardial I-R arrhythmia and shorten myocardial MIS, Therefore,apigenin plays a protective role on myocardial ischemia reperfusion injury.Apigenin can increase myocardial SOD activity and decrease the contents of myocardial MDA and serum LDH.The mechanism of protective myocardial ischemia reperfusion injury may be involved in the antioxidation.
