Objective:To explore the relationship between the expression of H-ras in Osteosarcoma and tumor pathological and clinical stage of Osteosarcoma.Methods:Expression of H-ras were determined in 63 Osteosarcoma tissues and 15 normal bone tissues with SABC in immunohistochemical method. Then analysis the posistive cells gray scale value by using image analysis system.And the expression of H-ras was also detect by Western blot in 4cases with Osteosarcoma tissues and their precancerous lesion tissues.Results: Both the immunohistochemical and the Western blot result showed that the H-ras expression in Osteosarcoma was much high then their precancerous lesion tissues or normal bone tissues.The positive rate of H-ras was 0% in the normal bone tissues ,While 42.85% in 63 Osteosarcoma tissues. The tumor pathological showed:the expression of H-ras in Osteosarcoma tissues was relevanted with infiltrate exent and lymphaticus aversion.There was had the different percentage of posistive case for H-ras in different clinical stage, in stage a1,2,3and4tumorswere0%(0/12),25%(2/8),52.94%(9/17),57.14%(8/14)and66.67%(7/12),respectively(p<0.005).Conclusions:The present data suggested that high expression of H-ras in Osteosarcoma tissues,and its espression was related to the tumor clinical stage and infiltrate exent and lymphaticus aversion.. It may be used as a maker of Osteosarcoma of the bone for the tumer stage,diagnosis and follow-up.
