The Significance Of Ezrin In Osteosarcoma

Objective : To investigate the expression Ezrin for osteosarcoma and the relationship between the positive rate of Ezrin and with lung metastasis-free,over survival time.Methods:Real-time polymerase chain reaction(PCR) was used to evaluate the levels of Ezrin,Six1,CD44mRNA expression in osteosarcoma from 39 treated patients and 16 cases of adjacent normal tissues ;the expression of Ezrin protein was detected in the tissue microarray which contains 56 spots of osteosarcoma tissue,10 spots of chondrosarcoma tissue and 10 spots of normal tissue by immunohistochemistry method.Next,we perform Western blotting to further investgate the expression in OS and normal tissues. The survival curve analysis and lung metastasis analysis with patients were evaluated through follow up.Results:The expression levels of Ezrin,Six1 mRNA were significantly higher in osteosarcoma than that in adjacent normal tissues. Although Six1 and CD44 genes have less significant influence on lung metastasis-free time and overall survival than Ezrin has. But high correlation with Ezrin gene was demonstrated in this experiment.The Pearson Correlation coefficients were 0.612 and 0.676, respectively. P value is 0.001 and less than 0.001.;Ezrin protein was also over-expressed in osteosarcoma tisues compared with that of chondrosarcoma and normal tissue.The positive rate of Ezrin in stage IIB osteosarcoma patients was 61.4 % , which was significantly lower than that of stage III patients (91. 7%) ( P < 0. 05)But it was showed not relationship between the positive rate of Ezrin and with gender,age,location,pathologic subtype and metastasis or not.Kaplan-meier analysis showed that patients with Ezrin positive expression had poor prognosis compared to patients with negative expression, however, negative patients had short time for lung metastasis . The specific Ezrin protein staining was observed in OS tissues and adjacent normal tissues. The expression level of Ezrin protein was significantly higher in OS than that in nomal tissues.Conclusion:The finding suggests that Real-time PCR is feasible method for detect Ezrin mRNA expression in OS; there was closely relationship between Ezrin and osteosarcoma associated with lung metastasis,Ezrin will become a useful evaluating marker for the prognosis and survival time of osteosarcoma .
