Objective: To observe the effect of Jin Shui Liu Jun Jian' s decoction on the content of hydroxyproline (Hyp) and the activity of Superoxide Dismutase (SOD) in both mice's skin and serum in the aged mice resulting from D-galactose, as well as the microstructural changes of the skin, and to partically infer Jin Shui Liu Jun Jian's function and mechanism of postponing the senility of skin.Materials and Methods: Kunming line mice, weighing 20-22g, were randomly divided into six groups (10 mice in each group): blank control group, aged model group, positive control group( Vitamin E 50mg/kg), low-dosage group (0.35g/ml), medium-dosage group (0.7g/ml) and high-dosage group (1.4g/ml), and they were gavaged respectively. The mice of blank control group were injected with 0.1ml/10g saline in the napesand backs every day, while other five groups were injected subcutaneously with 0.1ml/10g 1000mg/kg D-galactose in the napes and backs, continuously for 42 days to establish the subacute aged models. At the same time, all the mice were given corresponding garage therapy with 0.2ml/10g Jin Shui Liu Jun Jian's decoction, Vitamin E suspension and saline. Six weeks later, both the changes of activity of SOD and the content of Hyp in mice's skin & serum in all groups were tested. Optical microscopic examination of the skin tissue of all the mice was also performed.Results:①Change of antioxidase index of successful model: Compared with the blank control group, the content of Hyp in skin & serum and the activity of SOD in serum decreased obviously in the mice of aged model group (P<0.05), which showed the model establishment was successful.②Effect of Jin Shui Liu Jun Jian on antioxidation index of the skin tissue and serum: Compared with the aged model group, the content of Hyp (both in serum and in skin) and the activity of SOD in serum in the mice treated with the medium-dosage and the high-dosage Jin Shui Liu Jun Jian decoction and the Vitamin E suspension increased remarkably(P<0.05). However, the mice treated with Jin Shui Liu Jun Jian decoction and Vitamin E suspension demonstrated lower level of the activity of SOD in the homogenate of skin tissue than the model group and the blank control group. The activity of SOD in the homogenate of skin tissue of the blank control group was similar to those of the aged model group.③Results of skin tissue section in the model mice: Under the microscope, it was found that skin became thinner, the amount of collagenous fibers in dermal significantly decreased and fiber lined up loosely. For the mice in Jin Shui Liu Jun Jian group of medium-dosage and high-dosage and the positive control group, the skins were found thicker in microscope, the amount of collagenous fibers in dermal increased and fiber lined up thickly. However, there was insignificant difference in mice of the low-dosage group and the aged model group.Conclusions:①The mice models with skin aging are established by subcutaneous injection of 1000mg/kg of D-galactose once daily for 42 days. The changes of morphologic and biochemical parameters in these models are analogous to those in human skin aging.②Jin Shui Liu Jun Jian probably strengthens the activity of SOD, increases the content of Hyp and the amount of collagenous fibers in dermal in the aged mice.
