Objective: Compared the influence of the treatment of clearing away theheat-evil and expelling superficial evils with that of the treatment of promoting bloodcirculation to remove blood stasis on blood vessel endothelium of type 2 diabetesmellitus rat to investigate the mechanism of the two methods acting in the preventionand cure of type 2 diabetes mellitus and vasculopathy and the important role pyretictoxicity play in the occurrence and development of type 2 diabetes mellitus andvasculoathy.Methods: 63 Wistar rats were divided into nine groups at random: normalgroup, model group, gliclazide group, high, middle, low dose of Potentilla DiscolourBunge groups, high, middle, low dose of radix Salviae miltiorrhizae groups, sevenrats each group. Normal group rats fed with normal stoyer, the rest groups rats werefed with high calorie, glucose and fat stoyer first for a month and treated withstreptozotocin (30mg/kg) by peritoneal injection twice in a week to establish themodel of type 2 diabetes mellitus a month later. Tested the 2h postprandial bloodglucose of rats, which exceed 11.1mmol/L is needed, which less than it treated withstreptozotocin (20mg/kg) again until its blood glucose is agree with the standard.When the model was successfully established, rats were fed with normal stoyerinstead and treated with drug. Model group rats were garaged with normal saline.Gliclazide group rats were garaged with 10mg/kg.d gliclazide. High, middle, low doseof Potentilla Discolour Bunge groups rats were garaged with 30g/kg.d, 15g/kg.d,7.5g/kg-d Potentilla Discolour Bunge respectively. High, middle, low dose of radixSalviae miltiorrhizae groups rats were garaged with 12g/kg.d, 6g/kg-d, 3g/kg.d radixSalviae miltiorrhizae respectively. The course of treatment was six weeks.②Observed the variation of rat's body weight in each group.③Observed the variationof rat's 2h postprandial blood glucose and serum insulin content in each group.④Observed the variation of serum endothelin content in each group.⑤Observed the change of ultrastructural of rat's abdominal aorta endothelium in each group withtransmission electron microscope.Results:①the variation of the rats weight: After treatment, the weight ofmodel group's rats is decreased obviously compared with pretherapy (P<0.01), whilethe weight of the rest groups is rose obviously (P<0.05).There is no significantdifference in weight balance between Potentilla Discolour Bunge groups andgliclazide group. The weight balance of radix Salviae miltiorrhizae groups is lowerthan Potentilla Discolour Bunge groups and gliclazide groups', and there is significantdifference (P<0.05).②the variation of the 2h postprandial blood glucose of rats: Thevariation is not obvious both in normal group and radix Salviae miltiorrhizae groups.And there is no significant difference in them. After treatment, the value of modelgroup's rats is increased obviously compared with pretherapy (P<0.01), while thevalue is decreased obviously compared with pretherapy both in gliclazide group andPotentilla Discolour Bunge groups (P<0.05). There is no significant difference in theblood glucose balance between gliclazide group and Potentilla Discolour Bungegroups. The blood glucose balance of radix Salviae miltiorrhizae groups is lower thangliclazide group and Potentilla Discolour Bunge groups, and there is extremesignificant difference (P<0.01).③The variation of rats' insulin in serum: Thevariation is not obvious in normal group compared with pretherapy. And there is nosignificant difference. After treatment, the serum insulin content of the radix Salviaemiltiorrhizae groups and model group's rats is decreased obviously compared withpretherapy (P<0.01). After treatment, the serum insulin content of the PotentillaDiscolour Bunge groups and gliclazide group's rats is increased obviously comparedwith pretherapy (P<0.05). There is no significant difference in the serum insulinbalance between the Potentilla Discolour Bunge groups and gliclazide group. There isextreme significant difference in the serum insulin balance between the radix Salviaemiltiorrhizae groups and the rest treat groups (P<0.01).④The variation of rats'insulin in serum: The variation is not obvious in normal group compared withpretherapy. And there is no significant difference. After treatment, the serumendothelin content of model group's rats is increased obviously compared with pretherapy (P<0.01), while the serum endothelin content of the rest groups isdecreased obviously compared with pretherapy (P<0.01). There is no significantdifference in the serum endothelin balance among the high dose of PotentillaDiscolour Bunge group, the middle dose of Potentilla Discolour Bunge group andgliclazide group. The serum endothelin balance of radix Salviae miltiorrhizae groupsand low dose of Potentilla Discolour Bunge group are lower than gliclazide group,and there is significant difference (P<0.05).⑤The ultrstructure of rat's abdominalaorta endothelium: The normal group is normal. The change in the model group isvisible. The injure in rats abdominal aorta endothelium of all the groups treated withdrug is lighter than the model group. The change in the gliclazide group, high dose ofPotentilla Discolour Bunge group, high dose of radix Salviae miltiorrhizae group arelightest. The change in the middle dose of Potentilla Discolour Bunge group and themiddle dose of radix Salviae miltiorrhizae group is visible. The change in the lowdose of Potentilla Discolour Bunge group and the low dose of radix Salviaemiltiorrhizae group is obviously visible.Conclusion: Successfully established the type 2 diabetes mellitus rat model.The rat's endothelium was injured which indicates that there maybe existvasculopathy. The treatment of clearing away the heat-evil and expelling superficialevils and the treatment of promoting blood circulation to remove blood stasis cansuppress the oversecretion of endothelin, so that it can protect the blood vesselendothelium from injured and body weight from decrease. The treatment of clearingaway the heat-evil and expelling superficial evils can also down-regulate the bloodglucose and improve the insulin secretion. The difference of each group suggest thatthe treatment of clearing away the heat-evil and expelling superficial evils is betterthan the treatment of promoting blood circulation to remove blood stasis in protectingthe diabetes mellitus vascular endothelial cells.The study indicates that the treatment of clearing away the heat-evil andexpelling superficial evils and the treatment of promoting blood circulation to removeblood stasis can prevent the occurrence and development of the diabetes mellitusvasculopathy. It reveals the mechanism of the two methods acting in the prevention and cure of type 2 diabetes mellitus and vasculopathy and the important role pyretictoxicity play in the occurrence and development of type 2 diabetes mellitus andvasculoathy.
