The coronary heart disease (CHD) becames a familiar and common diseasewhich has threatened the adult and elderly people seriously. Percutaneous coronaryintervention (PCI) used widely as the most important technique in the therapy of theartery stenosis because it is safety and effective. However, the high rate of restenosis(RS) after PCI emerged more and more, this limit its application and development inthe clinical practice. As a result, it is a hard work to find a way to prevent the restenosisafter PCI. At the same time, the Traditional Chinese Medicine(TCM) recognized thatRS after PCI belongs to the category of "stagnation of blood symptom", the maincharacteristics is Xiongbixintongbing and the catheter's damage. The basicpathogenesis of RS is Benxvbiaoshi, the main feature of Benxv is deficiency of Zangqi,and the major feature of Biaoshi is stagnation of blood, so supplementing Qi andpromoting blood circulation are the basic therapeutic principle for preventing RS afterPCI. We selected a compound prescription by three kinds of herbs (Sanqi, Huangqi, andChuanxiong) which have effective function of reinforce Qi and promote bloodcirculation.Objective:Based on our ever research, we demonstrated the Yiqihuoxuefang caneffectively inhibit the neointimal proliferation induced by balloon injury on carotid ofbig rat, the possible reason is it can intervene the process of the cell cycle regulatorexpression. In oder to know about the herb also can effect the changes of theextracellular matrix (ECM), we observed the expression of the collage (typeⅠand typeⅢ) and the (MMP2) at the injury artery, to discuss the more mechanism of theYiqihuoxuefang. Morover, the compare between the compound prescription ofYiqihuoxuefang and the single herb of Huangqi was perfomed.Methods:60 male Sprague-Dawley big rats were randomly divided into 5 groups: thesham-operated group(A), the model group (B), the Huangqi group (C), the low-doseYiqihuoxuefang group (D), and the high-dose Yiqihuoxuefang group (E). Each groupinclude 2 time points (5day and 28 day ,n=6), the balloon catheter of 2F Fogarty wasperformed on the left common carotid artery of big rat ,the expression of the mmp2 was observed at two time points and the collage of typeⅠ,typeⅢwas observed at 28 daysafter injury by immunohistochemical technique.Results:All of the groups with herbs inhibit the neointimal proliferation significantly.Yiqihuoxuefang and Huangqi can suppress the expression of MMP2 at the 5th day afterballoon injury(p<0.01), the low and high dose Yiqihuoxuefang group are the best effect(p<0.01), but there are no statistical relationship between quantity and effect (p>0.05).the Huangqi group is obviously werker than Yiqihuoxuefang group (p<0.05); at the 28days after balloon injury, the expression ofmmp2 is not obvious, there are no statisticalrelationship between all of 5 groups. Yiqihuoxuefang and Huangqi can suppress theexpression of the collage (typeⅠand typeⅢ) at the 28th day after ballooninjury(p<0.01), the low and high dose Yiqihuoxuefang group are the best effect(p<0.01), but there are no statistical relationship between quantity and effect (p>0.05),the Huangqi group is the weakest in all groups of traditional herbs. (p<0.05).Conclusions:Yiqihuoxuefang and Huangqi can inhibite the proliferation of extracellular matrix.Yiqihuoxuefang can suppress the expression of MMP2 at the 5 days after the ballooninjury, on the other hand, ~iqihuoxuefang can suppress the expression of collage (typeⅠand typeⅢ), the high-dose and low-dose Yiqihuoxuefang have the more effective thanHuangqi (p<0.01), but there are no statistical relationship between quantity and effect(p>0.05). Yiqihuoxuefang and Huangqi can inhibite the proliferation of extracellularmatrix not only due to decreasing the expression of MMP2, but also suppressing the theexpression of collage (typeⅠand typeⅢ), while it is well proved that the compoundtraditional herb of Yiqihuoxuefang has a better effect than single herb of Huangqi.
