Objective: Basing on TCM Theory,the rats model of phlegm anti-blood vessels inthis subject was established by giving high fat diet.To obseverve themanifestation, the changes of aortic pathomorphism and blood lipid so as toevaluate the success of the model.The cellular and molecular mechanismabout pathogenic of phlegm evil was probed though the expression detectionof NF-κB.Observed the effect of GBHTTMR on the model in order todisprove whether the model is successful,and meanwhile clarify theanti-inflammatory and immunoregulation mechanisms.Methods: Fifty normal male Wistar rats of seven weeks old and 200±20g wererandomly divided into five groups:normal control group,model group,highdosage GBHTTMR-treated group(high dosage group), low dosageGBHTTMR treated group (low dosage group) and western medicine controlgroup; Observed the general circumstance of the big rats, including thechanges of food quantity, activity, hair glossiness and so on. Observed thecontents of TC, TG, HDL and LDL of rats' blood plasma in each group aftersentencing to death. Used immuno histochemical stain to examine theexpression of NF-κB in ascending aorta endothelium,then the results wereanalyzed by picture fixed amount. Observed the ultrastructural pathologicalstates of macrophage by using transmission electron microcope.Results: The rats in model group had the appearance of poor appetite,lack ofdoing,back arching,lethargy, yellow and lackluster wool,and hair dropping,etc.The big rats in normal group lively actived,and showed luminous of the woolcolor, the appetite were exuberant. The blood fats horizontal between modelgroup and normal group had the remarkable difference (P<0.01); After thetreatment, TC,,TG,LDL-c reduced and HDL-c elevated. TG, LDL-c in lowgroup was lower than model group; HDL-c was elevated and had statistical difference (P<0.05);TC was higer than model group with significantdifference (P<0.01). High dosage group and western medicine control grouphad the obvious difference with it (P<0.01); Compared with western medicinecontrol group, TG in high dosage group had the obvious difference (P<0.05),But others did not; TC, HDL-c, and LDL-c in Low dosage group hadremarkable difference (P<0.01), But TG did not. Compared with low dosagegroup, TC, LDL-c in high dosage group had the difference firmly (P<0.05),TG, HDL-c had the remarkable difference (P<0.01). The ultrastructure resultshowed that in model group blood intima was adema,broadening, evendiscontinuous and incomplete under blood vessel bast. In the blood themonocyte and the lymphocyte infiltrated to intension board. Manymacrophages could befound and there were massive all levels of dissolve theenzyme body and swallow slightly soak in its cytoblastema,The endosarc netand the ribose body was increased and outside the amnion the pseudopodappeared. Endothelial cell morphogenesis changed. It is obvious thatmitochondria was dropsy, cytoblastema was non-uniformity. After thetreatment of Chinese medicine treatment, above pathology harm obviouslyreduced. The expression of NF-κB in model group elevated, and had theremarkable difference comparing with normal group(P<0.01),and expressesafter the treatment reduced, it had the remarkable difference (P<0.01)comparing firmly with high dosage group.Conclusion: Through observed the expression of NF-κB in aorta bast, Our findingssuggest that the increased activation of NF-κB in aorta bast played animportant role in the pathogenesis of phlegm disease. The functionalmechanism might be that NF-κB was meditated the pathogenesis course bycontrolling the expression of cytokine,adhension molecule and inflammatorymediators.The mechanism was probably related to the reduce of blood lipidand damage of hyperlipemia on. vein endodermis, and also was connectedwith the activation of endodermis and maintaining the normal structure of vein endodermis and endothelial cells. It could delay and prevent the developmentfrom hyperlipemia to AS by inhibiting the activation of NF-κB andexpression of adhension molecules such as ICAM-1,VCAM-1,E-selectin andP-selectin as well as the adhesion ofmonocytes and endothelial cells.
