Asarum is root and rootstock of Asarum heterotropoides Fr.vat.mandshuricum(Maxim.) Kitaga.,Asarum sieboldii Mip.var.seoulense Nakai and Asarum rieboldii Mip.We called Asarum heterotropoides Fr.var.mandshuricum(Maxim.)Kitaga and Asarum sieboldii Mip. var.seoulense Nakai as Asarum heterotropoides Fr.var.mandshuricum(Maxim.)Kitaga commonly,It is a traditional chinese medicine which has been recorded in the edition 2005 of Pharmacopoeia of the PRC for treating cold,cough,headache,et al.12 Xixin samples were collected from different habitats,which were used for the studies on the chemical components, the method of assessment and antibacterial activities.The dried root and rootstock of Xixin was extracted with 70%ethanol.By solvent extraction,various chromatographic techniques of isolation(Si gel CC,PTLC,Sephadex LH-20,Semi-preparative Chromatography)and recrystallization,4 compouds were isolated and purified form CHCl3 extract.By means of spectroscopic analysis and physiochemical properties,3 of them were identified as:L-asarinin,L-sesamin,kakuol,daucasterol andβ-sitosterol.A RP-HPLC method was developed for simultaneous determination of 3 compounds in Asarum heterotropoides Fr.Schimidi var.mandshuricum(Maxim.)Kitaga..A Diamonsil C18 column was used with mobile phase being acetonitrile -water system,isocratic elution and the detection wavelength was set at 230 nm.The linear ranges for L-asarinin,L-sesamin and kakuol were 3.50-35.0μg·mL-1(r =0.9997);1.52-76.0μg·mL-1(r =0.9992);0.38-7.60μg·mL-1(r =0.9999),respectively.The average recovery of L-asarinin,L-sesamin and kakuol were 99.7% (RSD = 2.7%),96.7%(RSD = 1.9%)and 97.7%(RSD = 1.5%),respectively.The assay method was simple,rapid,reliable and reproducible and provided a quantitative basis for the quality assessment of Xixin.The orthogonal design was adopted to optimize the extraction of L-asarinin,L-sesamin and kakuol.The optional extracting condition was methaol consumed ten times the amount of material,and there times for 20 min each time.Paper method and 96 shadow mask method were used to detect the antibacterial activities of L-asarinin,L-sesamin and kakuol on Escherichia coli(EC),Staphylococcus aureus(SA), pneumonia(PN),Pseudomonas aeruginosa(PAK)and Candida albicans(CA),and the MIC of L-asarinin,L-sesamin and kakuol on the five becterias were detected,respectively.The 96 shadow mask method consumed little medicine,and has a result which could easliy be deteceed and provided a pharmacodynamic basis for the study of Xixin.
