Objective: Parkison's disease (PD), which is a parapymmid progressive neurodegenerative disease in centre nerve systerm., always invades middle-ages and older person and causes their disables. PD's mainly pathology change maintains the death of most of the dopamine neurons which from tight zone of substantial nigra of midbrain to striatum. PD's mainly symptoms include stilly shacking, bradykinesia, rigidity muscles and abnormal posture and pace. During these years the rate of PD is climbing. It is recognized that PD is hard to be cure. In the world, Nowadays, western medicine could not prevent the progress of PD well, but bring about a lot of side effects at the meantime. Operation and gene therapy could not treat the PD well either. At present, acupuncture as a peculiar, safety, and convenient way has been applied in the treatment of PD and has good affection. The experiment will study the rats' rotation and the quantity of TH,BDNF,TrkB and CREB in order to study the therapeutically effect and mechanism of the electroacupuncture (EA) on Parkinson's disease(PD), which will offer experimental basis of applying EA in clinic practice of the acupuncture-moxibustion.Methods: The Parkinsonian rats were modeled by stereotaxieal microquantitative injection in the right striatum with 6-hydroxydopamine and treated by the electroacupuncture therapy of Fengfu. All the experimental rats were divided into four groups: Normal Group, Sham-modeling Group, Model Group, Electroacupuncture Group. (1) Observation on behaviors: apomorphine was injected into abdominal cavity to induce left rotation. The number of circles rotated within 30 min after injection of apomorphine was recorded. (2) Histochemical and immunohistoehemical staining methods area adopted to observe the morphologic changes of substantia nigra and quantify the tyrosine hydroxylase (TH) positive neurons by the immunohistochemistry. (3) The therapy effects of the the BDNF,TrkB,CREB positive cells in substantia nigra.Result: (1) In EA group, the number of rotations after the treatment was lower than before the treatment(P<0.05), but that didn't change in PD group(P>0.05). (2) The number of TH in PD group was obviously lower than that of the normal group(P<0.01), and that in the EA group was higher than the PD group(P<0.05). (3) the number of BDNF, TrkB and CREB positive neurons In PD group was obviously less than those of the normal group and sham-operation group (P<0.01).In the EA group, the number of BDNF, TrkB and CREB positive neurons were more than that of the PD group(P<0.05).Conclusion: BDNF plays a very important role in protective DA neurons and maintenance their normal function, demonstrates the attractive prospect in the PD treatment. EA may obviously prevent the PD mouse to revolve. Its possible mechanism promising:(1) Increases the TH content, is advantageous forms L-Dopa in the L-tyrosine hydroxy, inereases DA to synthesize; (2) Enhance the contents of BDNF, which can promote the regeneration of DA neurons. (3) Enhance the contents of BDNF, TrkB in order to stimulate the CREB which is in karyon, thus enhances BDNF in the opsoninum from the secretion, protects DA neurons.
