Objective:To observe the effects of Shenshuai-Ⅰdecoction on expression of angiotensinⅡ(Ang-Ⅱ)in kidney of unilaternal ureteral obstruction(UUO)rats,and explore the mechanism of the effect.Methods:60 Wistar rats were randomly divided into five groups:model group,Shenshuai-Ⅰgroup,Benazepril group,sham operation group,normal group.12 rats were in every group. Inject 30mg/kg 2%Pentoarbital Sodium in abdominal cavities of the rats of model group,Shenshuai-Ⅰgroup,Benazepril group and sham operation group,and fixed them after they were anesthetized.Prepare skin;opened abdominal cavities;ligatured left ureters and sewed up the wounds.Every day gave every rat an intramuscular injection of 80 thousand units penicillin to prevent infection.Every day poured 10ml/kg physiology salt solution into the rats'stomachs of model group in 14 days. Shenshuai-Ⅰgroup:every day poured stomachs with Shenshuai-Ⅰdecoction in 14 days,and dose was 8g/(kg.d). Benazepril group:every day poured stomachs once in 14 days,and dose was 5mg/(kg·d).Sham operation group:samely anesthetized and opened abdominal cavities,but didn't ligature ureters;one layer by one layer sewed up the wounds,gave an intramuscular injection of penicillin,every day poured stomachs with 10ml/kg physiology salt solution in 14 days.Normal group:didn't perform operation;only gave an intramuscular injection of penicillin in three days and poured stomachs with 10ml/kg physiology salt solution in 14 days.Adopted randomly 10 rats from every group into metabolic cages from the thirteenth day after ligation of ureters to gather 24 hours urine,and delivered it to Hubei Province Traditional Medicine Hospital to measure theβ2-mG content in urine.Excise eyes of every group rats on the fourteenth day,and gathered blood into test tubes.Delivered the blood to the Chechout Room of Hubei Province Traditional Medicine Hospital to measure the creatinine content.Made the roots died;got the kidneys of ligation side and preserved them into 10%neutral formalin to make kidney tissue into slices and measure the angiotensin-Ⅱcontent in them.After making kidney tissue into slices and being dyed by Hematoxylin and Eosin,selected randomly ten slices from every group to observe and compare degree of renal interstitial fibrosis of various groups under high times lens(×200).Appraise the result adopting one-sided blind method.Scored it by Priraini method:divided into four grades(-,+,++,+++),and respectively kept the score with "one,two,three,four".(-):renal tubules and interstitium hadn't pathological changes.(+):pathological changes scope was smaller than 20%.(++):pathological changes scope was from 20%to 40%.(+++):pathological changes scope was bigger than 40%.Continuously observed ten repeatedless high times fields of vision,and synthesizly gave marks.Adopted SABC method to determine content of expression of angiotensin-Ⅱin kidney tissue.Selected randomly ten slices from every group,and analyse by HPIAS2000 Picture Analyce Software form Tongji Qianping Portrait Company.Adopted randomly five disoverlapping fields of vision from every slice under high times lens(×400),and positive dyeing parts were brown.Measure average luminosity of dyeing parts.Statistical method:analysed the data by SPSS11.5 software. Quantification data was expressed to(?)±S,and compared every group by T-test.Result:1.Measure ofβ2-mG content in urineβ2-mG content in urine in Shenshuai-Ⅰgroup and Benazepril group was obviously lower than it in model group(p<0.01),and was higher than it in sham operation group and normal group(p<0.01).Hadn't obviously diffence in Shenshuai-Ⅰgroup and Benazepril group(p>0.05).2.Measure of creatinine content in bloodShenshuai-Ⅰgroup and Benazepril group hadn't statistical difference from sham operation group and normal group(p>0.05).Shenshuai-Ⅰgroup and Benazepril group hadn't statistical difference from model group(p>0.05).3.Pathology observationHE dyeing pointed out that structure of kidney tubules was normal of sham operation group and normal group.Part of kidney tubules of model group dislated;tubule cortex became thiner;part of kidney tubules were atrophied and inflammatory cells obviously infiltrated.Pathological changes of Shenshuai-Ⅰgroup and Benazepril group were slighter than model group.Hadn't obviously diffence in Shenshuai-Ⅰgroup and Benazepril group.4.Score of HE dyeing of renal interstitial fibrosisScore of Shenshuai-Ⅰgroup and Benazepril group was obviously lower than model group(p<0.01),and was taller than sham operation group and normal group(p<0.01).Score of Shenshuai-Ⅰgroup was equivalent to Benazepril group(p>0.05).5.Positive expression of angiotensin-Ⅱin kidney tissueAverage luminosity of Shenshuai-Ⅰgroup and Benazepril group was obviously lower than model group(p<0.01),and higher than sham operation group and normal group(p<0.01). Average luminosity of Shenshuai-Ⅰgroup was lower than Benazepril group(p<0.05).Conclusion:The research indicated that Shenshuai-Ⅰdecoction has effect of preventing renal interstitial fibrosis.One of the effect mechanisms probably be to prevent expression of angiotensin-Ⅱin kidney tissue,and to prevent angiotensin-Ⅱ's effect in renal fihrosis:①guided proliferation and hypertrophy of interstitial fibroblast;②led to gather of extracelluarmatrix;③effect of inflammation medium;④promotive effect to lower reaches factors to guide renal fibrosis.
