The optimal extraction process of "Chelidonium majus L "was studied by orthogonal design with extractive yield and the contents of sanguinarine, berberine and chelerythrine as indices.Four factors were investigated in this experiment,including the quantitation of solvent, time of extraction.,soak time,times of extraction.The results showed that the optimal extracting condition was 85% ethanol consumpted 6 times the amount of material, refluxing for twice and 0.75hour per time.RP-HPLC methods were developed to determine sanguinarine, berberine,chelerythrine and protopine in "Chelidonium majus L". To determine the content of sanguinarine,berbernine and chelerythrine in Chelidonium majus. simultaneously,the HPLC with a Diamonsil C18 (250 mm×4.6mm,5μm) ,was used.Acetonitrile-1 % triethylamine(25:75 adjusted to pH3 with phosphoric acid) was used as mobile phase at the flow rate of 1mL·min-1,the detection wavelength was at 263nm. Assay was linear over the range 6.0-120μg·mL-1 for sanguinarine (r=0.9996), 3.0 - 60μg·mL-1μg·mL-1 for berberine (r=0.9999), 10.0-200μg·mL-1 for chelerythrine (r=0.9998). Average recoveries of samples were 99.2%(RSD 1.8%), 102.0% (RSD 2.0 %), and 98.14% (RSD 1.9 %) for three constituents, respectively. To determine the content of protopine in Chelidonium majus.,the HPLC with a Diamonsil C18 (250mm×4.6mm,5μm) ,was used.Methonal-1%triethylamine(32:68) was used as mobile phase at the flow rate of 1mL·min-1,the detection wavelength was at 285nm. Assay was linear over the range 0.5-12μg·mL-1 for sanguinarine (r=0.9995), average recovery of sample was 97.8%(RSD 1.7 %).The assay methods are simple,rapid and reproducible,which provide a quantitative basis for the quality assessment for "Chelidonium majus L ". The residues of Cu, Cd ,As, Hg, and Pb in samples are in the limit of the standard.To investigate the stability of these methods,the contents of sanguinarine,berbernine and protopine in "Weitongping syrup" were achieved.The HPLC analysis was accomplished as above.A better linearity and good correlation coefficient were obtained: 5.92-118.4μg·mL-1 of sanguinarine (r=0.9999),4.0-80.0μg·mL-1 of berbernine (r=1.0000),2.0-100.0μg·mL-1 of protopine (r=0.9997) .The average recoveries were obtained:97.9 % for sanguinarine(RSD 1.8%), 97.7% for berbernine (RSD 1.0%),101.2% for protopine (RSD 1.0%). The assay methods are simple,rapid and reproducible,which provide a quantitative basis for the quality assessment for "Weitongping syrup".
