Through ligating unilateral ureteral of the rats,we can observe the effect of jiajiancaoguozhimutang decoction on expression of CTGF in kidney of unilaternal ureteral obstruction(UUO)rats,further explore its possible role in interstitial fibrosis mechanism.Methods:1.30 rats is SPF level and healthy adult male rats(150g-200g),they were randomly divided into the model group(n=6),jiajiancaoguozhimutang group(n=6),benazepril group(n=6),sham-operated group(n=6),the blank group(n=6).2.We ligate the left ureteral of thirty rats to make interstitial fibrosis model.The left ureteral of rats were ligated in the model group, jiajiancaoguozhimutang group,benazepril group.Sham operation group are dressed after skin incision.Blank group are not operated.Penicillin 80,000 units intramuscular injection per day three days from the ligation date is to prevent infection.3.Model group:the rats are infused with saline,3 ml daily. Benazepril group:the rats are gived with benazepril,10 mg/g daily. Jiajiancaoguozhimutang group:the rats are gived with jiajiancaoguozhimutang,3g/ml daily.Sham operation group:the rats are dressed after skin incision and are infused with saline,3 ml daily.Blank group:the rats are infused with saline only,3 ml daily.To 13 days, collect specimens.4.By observing the changes of urineβ2-microglobulin and serum creatinine,we understand jiajiancaoguozhimutang on the role of UUO rats. Immunohistochemistry was used to detect connective tissue growth factor expression in each group of renal obstruction,by pathological section of renal tissue we can observe changes in renal structure of the organization and the pathology integral of tubulointerstitial as indicators to observe effect of jiajiancaoguozhimutang on the UUO rat renal interstitial fibrosis we can observe.Results:1.General performance:jiajiancaoguozhimutang groups and model groups and benazepril groups:the increasing size of the obstructed kidneys with prolonged obstruction,renal stagnant water,thinning renal parenchyma,compressing renal pelvis nipple,bending fornix parts nipple, some nipple ischemic and atrophy,effusion transparent,and the renal subcapsullar spot hemorrhage.2.HE staining showed that renal tubular structure is normal in the blank group and sham-operated group,model group of tubular lumen expansion,tubular epithelial cells were flat,some tubular atrophy, inflammatory cell infiltration,jiajiancaoguozhimutang group and benazepril group lesions is less than the model group.According the HE staining,the pathological scores of the interstitial were also significantly higher in the model group than jiajiancaoguozhimutang group and benazepril group,significantly higher in the m3.Two weeks later,urineβ2-MG content were significantly higher in model group than the sham-operated group and the control group(P<0.01), urineβ2-MG content was significantly lower in jiajiancaoguozhimutang group and benazepril group than model group(P<0.01),urineβ2-MG content in jiajiancaoguozhimutang group and benazepril group,there were no significant difference(P>0.05).4.Connective tissue growth factor expression was significantly lower in jiajiancaoguozhimutang group than the model group(P<0.01). Connective tissue growth factor expression was significantly high in jiajiancaoguozhimutang group than the benazepril group(P<0.01).5.There are not statistical significance between each group about Scr content.Conclusion:This study shows that:UUO rat kidney and renal tubular was damaged mainly in the interstitial fibrosis.The over expression of CTGF may be one of the mechanisms of renal fibrosis.Jiajiancaoguozhimutang can reduce urinaryβ2-MG content and the expression of CTGF,and resistance the injury of UUO rats.
