Effect Of Liuweidihuang Decoction On Expression Of Connective Tissue Growth Factor And α-Smooth Muscle Actin In SD Rats With Unilateral Ureteral Obstruction

With Sprapue-Dewley rat models with UUO, to examine the effects of Liuweidihuang decoction on the renal function, pathologic changes of the obstructive kidney, expression of CTGF andα-SMA in the kidney.Results:1.The effect of YISHEN decoction on the pathologic changes of structure in obstructive kidney of rats with UUO. After operation,the operated rats showed progressive renal interstitium fibrosis.The tubulointerstitial fibrosis was more severe in UUO groups than that in Chinese herbs group and the time depending was found more extracelluar matrix(ECM).2.The effect ofLIUWEIDIHUANG decoction on the expression of connective tissue growth factor(CTGF) in rats with UUO.The degree of CTGF expression were allayed by LIUWEIDIHUANG decoction treatment.In the experiment, with Immunohistochemistry method we find that there is more light dyeing and smaller stained area of CTGF in obstructive kidney of LIUWEIDIHUANG therapy groups than Sham groups.But there was no significantly between LIUWEIDIHUANG group and Enalapril group.3.The expression ofα-smooth muscle actin, In Sham groups,noα-SMA was detected in tubulointerstitium,except that there was staining ofα-SMA in the vascular smooth muscle cells.In contrast, there was strong staining ofα-SMA within tubulointerstitium in UUO groups.but In comparison with the rats of operation group,the Liuweidihuang decoction treated rats had lessα-smooth muscle actin in interstitium.Conclusion:our results shows that CTGF may plays a key role in process of tubulointerstitial fibrosis. Liuweidihuang decoction have the effect of down-regulation on CTGF andα-SMA therefore decrease renal tubulointerstitial fibrosis.
