Objective: To explore whether homocysteine(Hcy) is an independent effect factor for nitric oxide synthetase (NOS) of penile corpus cavernosum of type 2 diabetic adult male Wister rats.Methods: Ten rats were randomly selected. from fifty Wistar masculine rats of 3 months supplied by normal feeding. as contral group(A . group ; n=10) . Other forty rats were fed by high-fat and high glucose . After 4 weeks they were injected in vena caudalis with streptozotocin (30mg/Kg,). Selected thirty diabetic rats divided into three groups: diabetic rats group (B group ; n=10). ; diabetic rats treatment with insulin group (C group ; n=10). ; diabetic rats treatment with folic acid and vitamin B12 group ( D group ; n= 10) ; A group supplied by normal feeding ; B,C,D groups were fed by high-fat and high glucose . Injected with apomorphine,penile erection of the rat models with DM and the controls were observed and noted on the 8th and 12th week .The level of the total plasma homocysteine were measured by method of cycle enzyme. NOS activity was examined in selected rat models.Results: 1. Compared with the controls, the level of plasma tHcy of B group was obviously increased . NOS activity in the penile cavernous tissue and the erection of rats with DM decreased significantly (P<0·01) and also decreased significantly(P<0·01) with the increased duration of DM. .2 . Compared with B group , The level of plasma tHcy of C group was obviously decreased .NOS activity in the penile cavernous tissue and the erection rate of C group increased significantly ( P < 0. 05) . 3 Compared with B group , The level of plasma tHcy of D group was obviously decreased. NOS activity in the penile cavernous tissue and the erection rate of D group increased significantly.( P < 0. 05) .Conclusions: The level of plasma tHcy of type 2 diabetic of rats is an independent effcet for nitric oxide of rat penile corpus cavernous of adult male Wister. Moreover,Hcy may constitute a new risk factor for DMED.
