ObjectiveTo research the influence of extracting solution of Hirudo on bovine retinal pigment epithelial(RPE) cells' signal transduction that is guided by protein tyrosine kinase(PTK). Methods(1) Five kinds of conventional extracting solution of Hirudo (alcohol and water extracting, alcohol refluxing, water extracting, water extracting-alcohol precipitating, percolation filtration) were used in this study. Bovine RPE cells that were cultured in vitro were exposed to different kinds of extracting solution in different concentrations for 24 hours. MTT method was used to detect the OD values, and the rates of inhibition were calculated. Selected the proper extracting solution and concentration mat inhibit the proliferation of RPE cells greatly.(2) Bovine RPE cells were exposed to solution of Platelet-derived growth factor(PDGF) with different concentrations for 24 hours. Got the OD values and the rates of proliferation. Selected the proper concentrations that proliferate RPE cells greatly.(3) The choosen 16mg/ml percolation filtration of Hirudo and 0.02 ug/ml PDGF were used to incubate RPE cells for 15 minutes with three different modes (adding extracting solution of Hirudo first, then solution of PDGF; adding solution of PDGF first, then extracting solution of Hirudo; adding the...
