Objective:Study the protective effect and mechanism of Puerarin against retinal damage by light.Methods:Examine the pathological changes of retina by light microscopy; and test TNF-α,FasL,NF-κB and Caspase-3 into retina by immunohistochemical technique.Results:(1) After the light exposure,the outer nucleus layer of retina in the model group became thinner and the regular arrangement of outer and inner segments were disordered,dividing line wasn't distinct.In the low-dose group the outer nucleus layer of retina got thicker than that of the model group.(2) the level of FasL,TNF-αand Caspase-3 in model group retina increased,NF-κB decreased.In low-dose group,the level of FasL,Caspase-3 and TNF-αobviously decreased,NF-κB increased.Compared with the model group,showed significant difference (P<0.01 or 0.05).Conclusion:Puerarin could release the photic injury of retina,accelerate the renovation of retina after light exposure and ameliorate the function of retina.The mechanisms might relate to improve NF-κB level and reduce FasL,TNF-αand Caspase-3 levels so as to inhibit the apoptosis.
